The Causal Relationship Between Gut Microbiota Circulating Inflammatory Factors and Scoliosis

Abstract

Objective: Based on Mendelian randomization MR analysis the causal relationship between gut microbiota GM), circulating inflammatory factors CIPs and Scoliosis SS was discussed. Methods: Data for 211 GM 91 CIPs and SS were obtained from the Genome-Wide Association Studies GWAS database. The inverse variance weighting IVW method was used as the main test in MR analysis. Sensitivity analysis was used to assess the robustness of MR results. Reverse MR Analysis was used to determine the directionality of causal relationships.Results: The results of the study showed that the levels of Eukaryotic translation initiation factor 4E-binding protein 1 and Caspase 8 in CIPs were positively correlated with SSOR>1P<0.05), whereas high levels of Osteoprotegerin and Transforming growth factor-alpha TGF-α were negatively correlated with SSOR<1P<0.05. In GMDesulfovibrio genus Eubacterium ventriosum group genus Lachnospiraceae FCS020 group and genus Ruminococcus 6 were positively correlated with SS OR>1P<0.05), whereas genus Bilophila genus Eubacterium eligens group and genus Prevotella 9 were negatively correlated with SS OR<1P<0.05. Heterogeneity and multiplicity tests confirmed the reliability of the MR analysis. Reverse MR analysis did not reveal that these risk and protective factors were reverse causally associated with SS. In addition the studies have found no mediating role of CIPs in the causal relationship between GM and SS.Conclusion: In CIPs 4E-BP1 and caspase 8 are risk factors for SS while osteophosphorin and TGF-α are protective factors for SS. In GMDesufovibrioEubacterium ventriosumLachnospiraceae FCS020 group and Ruminococcus 6 are the risk factors of SS while BilophiliaEubacterium and Prevotella are the protective factors of SS. This study provides new directions and targets for the diagnosis treatment and prevention strategies of SS.
