Abstract: Objective：To investigate the effects of medicated serum of Jiawei Xiaochaihu Decoction on the proliferation of hepatic stellate cells（HSC） and expression of connective tissue growth factor (CTGF) in rats.Methods：①Preparation of serum containing drugs：Wistar rats were randomly divided into high dose group， medium dose group， small dosage group and control group， each of 10 rats.Each group was given distilled water or the corresponding dose of Jiawei Xiaochaihu Decoction， 1 time a day，for 5 d. After the last administration of 2 h， blood samples were taken from the abdominal aorta to prepare the corresponding dose serum.②The effects of medicated serum of Jiawei Xiaochaihu Decoction on the proliferation of rHSC-99 cells and expression of CTGF： rHSC-99 cells near confluent growth were divided into high dose group， medium dose group， small dosage group and control group after digestion with 0.1% trypsin，and seeded in culture plate. After 24 h and 48 h culture， the effects of serum containing drugs on the proliferation of rHSC-99 cell line were detected by MTT， and the expression of cytokine CTGF and the expression of CTGF mRNA were detected by immunocytochemical staining and RT-PCR.Results：The serum containing different concentrations of Jiawei Xiaochaihu can inhibit the proliferation of HSC， and significantly inhibit the expression of CTGF protein and CTGF mRNA.Conclusion：Jiawei Xiaochaihu Decoction can inhibit liver fibrosis by inhibiting proliferation and inhibiting the expression of CTGF in HSC.
