放疗对非小细胞肺癌患者循环内皮祖细胞数量和功能的影响

Effect of Radiotherapy on the Number and Function of Circulating Endothelial Progenitor Cells in the Patients with Non-small Cell Lung Cancer

摘要: 目的:检测非小细胞肺癌(non-small cell lung cancer,NSCLC)患者放疗过程中循环内皮祖细胞(circulating endothelial progenitor cells,cEPCs)数量和功能的变化,探讨放疗对cEPCs数量和功能的影响。方法:选取2015年1月至2016年5月于我院住院确诊NSCLC并选择放疗患者30例为治疗组,以同期健康体检者30例为对照组。治疗组放疗前、放疗后第15天、放疗后第30天,对照组于入组后次日抽取外周静脉血分离单个核细胞,体外培养后鉴定内皮祖细胞,并检测cEPCs数量、迁移、粘附能力。结果:NSCLC患者放疗前cEPCs数量高于对照组;cEPCs迁移、粘附能力均高于对照组,差异有统计学意义(P<0.01);NSCLC患者cEPCs的数量及迁移、粘附能力在放疗后各期与放疗前的差异有统计学意义(P<0.05);治疗组cEPCs的数量在放疗后第30天最少,差异有统计学意义(P<0.05);治疗组cEPCs的迁移、粘附能力在放疗后第30天最低,差异有统计学意义(P<0.05)。结论:NSCLC患者cEPCs数量升高,迁移、粘附能力增强,放疗后cEPCs数量降低,迁移、粘附能力下降,检测cEPCs有可能作为NSCLC患者辅助诊断和评价治疗效果的指标之一。

Abstract: Objective:Changes on the number and function of circulating endothelial progenitor cells(cEPCs)after radiotherapy in patients with non-small cell lung cancer(NSCLC)were examined to investigate the impact of radiotherapy on cEPCs.Methods:Thirty patients from January 2015 to May 2016 in our hospital with NSCLC were enrolled as the treatment group, and thirty cases of health subjects as the control group at the same time. Peripheral venous blood samples of the treatment group were obtained before radiotherapy, fifteen and thirty days after radiotherapy respectively, and the samples of the control group were obtained the next day after admission. The number and function of cEPCs pre- and post-treatment were measured by density gradient centrifugation method.Results: The number of cEPCs significantly increased in patients with NSCLC compared with control subjects, and their cEPCs function of migration and adhesion ability enhanced, and the difference was statistically significant (P<0.01). In addition, cEPCs number and function of patients with NSCLC were decresed after radiotherapy, and the difference was statistically significant(P<0.05). The number and function(migration and adhesion ability) of cEPCs were decreased more thirty days after radiotherapy, and the difference was statistically significant(P<0.05). Conclusion:Radiotherapy can reduce the number of cEPCs and the migration and adhesion ability of cEPCs in patients with NSCLC.The detection of cEPCs may be one of the indicators for the adjuvant diagnosis and therapeutic effect of NSCLC patients.
