Expression and Significance of α-Synuclein in Rats with Spinal Cord Injury

摘要: 目的:探究大鼠脊髓损伤(spinal cord injury,SCI)后α-突触核蛋白 (α-synuclein, SNCA) 在脊髓中的表达和意义。方法:SD大鼠随机分为对照组和SCI组,采用Allen′s法制备大鼠SCI模型。根据基因芯片分析结果,筛选出目的基因。应用实时定量PCR和免疫组织化学方法分别检测SNCA在脊髓中的定位表达和变化。生物信息学分析预测SNCA相关基因或蛋白。结果:基因芯片结果显示差异下调基因SNCA差异表达倍数2.276,验证结果显示SNCA下调。实时定量PCR显示SNCA的 mRNA在大鼠SCI后3、7、14和28 d的表达量明显降低,与对照组相比,差异均有统计学意义(P<0.001);免疫组织化学结果显示SNCA在正常脊髓后角的神经元突起、神经胶质细胞突起以及前角神经元胞浆中表达,在SCI后7 d和14 d,与对照组相比,SNCA免疫阳性物质明显减少。生物信息学分析结果显示SNCA与神经递质传递、氧化应激和细胞凋亡等有关。结论:SCI后SNCA的mRNA和蛋白表达水平均降低,在脊髓前角和后角的表达明显减少。提示在SCI后SNCA可能参与调控神经元和神经胶质细胞的存活,进一步影响SCI后的运动功能和感觉功能。

Abstract: Objective:To investigate the expression and significance of α-synuclein (SNCA) following acute spinal cord injury(SCI) in rats. Methods:Rats were randomly divided into two groups which contain control group and SCI group.Rat model of contusion SCI was established with modified Allen′s method. Based on the results of gene array, the target gene was chosen. Real-time quantitative polymerase chain reaction (qRT-PCR) and immunohistochemistry staining were used to detected the level of SNCA expression and distribution in spinal cord.The SNCA related genes or proteins were predicted by Bioinformatics analysis. Results:Gene array results showed that the fold change of SNCA was 2.276 in down-regulated different genes,and the result of verifying was SNCA decreased.qRT-PCR showed that the level of SNCA mRNA was significantly decreased at 3,7,14,28 d after SCI(P<0.001).In addition,immunohistochemistry staining showed SNCA was found in the cytoplasm of neuron, axon and dendrite of glia at anterior horn or posterior horn of normal spinal cord.The density of immune-positive production were significantly decreased at 7d and 14 d after SCI when compared to control group.Bioinformatics analysis showed that SNCA was related to neurotransmitter delivery, oxidative stress and apoptosis. Conclusion:This study found that the level of SNCA mRNA and protein were reduced in rats with SCI, and the expression of SNCA in anterior horn or posterior horn of spinal cord was decreased after SCI.Results indicated that SNCA might be associated with the neuron or glia survival, which effect the motor function and sensory function after SCI.
