摘要: 目的:建立高效液相色谱(HPLC)法测定桑黄发酵菌丝体粉中麦角甾醇的含量。方法:采用甲醇超声提取菌粉中的麦角甾醇,色谱柱为Welch Mltimate LP-C18柱(4.6 mm×250 mm,5 μm),流动相为甲醇∶水(98∶2),流速1 mL·min-1,检测波长 283 nm,柱温30 ℃。结果:在进样量为0.100 721~1.007 214 μg(r2=0.999 9)范围内,麦角甾醇的浓度与峰面积具有良好的线性关系;平均加样回收率为98.02% (RSD 0.80%)。结论:该检测方法准确可靠,重复性好,可操作性强,可用于深层液体发酵桑黄菌丝体粉中麦角甾醇的含量测定,同时也为麦角甾醇列入桑黄发酵菌丝体粉的质量标准提供依据。

Abstract: Objective:To establish a HPLC method for the determination of ergosterol in deep fermented mycelium powder of Phellinus igniarius. Methods: Using methanol to extract ergosterol from mycelium powdeer by rultrasound, the chromatographic column was Welch Μltimate LP-C18(4.6 mm×250 mm, 5 μm)with methanol∶water(98∶2) as the mobile phase at a flow rate of 1 mL·min-1. The detection wavelength was 283 nm, and the column temperature was 30 ℃. Results:There was a good linear relationship between concentration and peak area within the range of 0.100 721-1.007 21 μg(r2=0.999 9). The average recovery rate was 98.02% (RSD 0.80% ). Conclusion:The method is accurate, reliable, easy operation and repeatable.It can be used to determine the content of ergosterol in deep fermented mycelium powder of Phellinus igniarius and also provide reference for ergosterol as the quality standards of Phellinus igniarius fermented mycelium powder.
