Determination of Active Ingredient Content in Jinyin Qingwen Mixture by HPLC

摘要: 目的:建立金银清瘟合剂中君药金银花主成分绿原酸含量测定方法。方法:采用水煎提取和蒸馏浓缩法提取处方中各药材有效成分制成合剂,采用HPLC法测定:Diamonsil G8 ODS色谱柱(4.6 nm×200 mm,5 μm);以50%甲醇-0.4%磷酸水溶液为流动相,流速1.0 mL?min-1,柱温30 ℃,检测波长327 nm,进样量10 μL。结果:绿原酸保留时间为18.126 min,线性范围为0.991~9.93(r=0.999 9),平均加样回收率为99.16%,检测限为0.1 μg?mL-1。结论:金银清瘟合剂制备工艺稳定,HPLC法测定绿原酸含量高效、准确、稳定,质量标准可行。

Abstract: Objective:To establish a method for the determination of chlorogenic acid content in jinyin qingwen mixture. Methods: Water decoction and distillation were used to extract the effective components of the medicinal materials. HPLC method was used to determine the content of chlorogenic acid, Use Diamonsil G8 ODS column (4.6 nm×200 mm, 5 μm), taking 50% methanol-0.4% phosphoric acid water as mobile phase,the flow rate was 1.0 mL?min-1, column temperature was 30 ℃, detection wave length was 327 nm, sample quantity was 10 μL. Results: Chlorogenic acid retention time was 18.126 min, linear range was 0.991-9.93(r=0.999 9), average recovery was 99.16%, detection limit was 0.1 μg?mL-1. Conclusion: The preparation process of jinyin qingwen mixture is stable.HPLC method for determination of chlorogenic acid content is efficient, accurate and stable, and the quality standard is feasible.
