Girdin 与VEGF在胶质瘤中的表达及其与血管生成的关系

Expressions of Girdin and VEGF in Human Glioma and Their Relationship to Angiogenesis

摘要: 目的:研究胶质瘤中Girdin与血管内皮细胞生长因子(VEGF)的表达及二者与胶质瘤血管生成的关系,探讨Girdin与VEGF在胶质瘤血管生成中的作用。方法:应用免疫组化Envision法分别检测Girdin和VEGF在正常脑组织和不同级别胶质瘤组织中的表达;同时应用CD34抗体标记胶质瘤血管内皮细胞,计数肿瘤微血管密度(MVD),并分析Girdin、VEGF及MVD三者之间的关系。结果:Girdin和VEGF在正常脑组织中均不表达,在胶质瘤中的表达均高于正常脑组织;随着胶质瘤WHO分级的升高,两者表达均呈升高趋势(P<0.05)。随着胶质瘤分级的升高,MVD计数呈上升趋势。高、低级别胶质瘤之间的MVD计数比较,差异有统计学意义(P<0.05)。胶质瘤中Girdin表达与MVD呈正相关(P<0.01),VEGF表达与MVD呈正相关(P<0.01),Girdin表达与VEGF表达亦呈正相关(P<0.01)。Girdin表达与胶质瘤KPS评分、切除范围和胶质瘤分级相关(P<0.05),与年龄、性别无相关性。结论:胶质瘤中Girdin和VEGF高表达,两者表达与胶质瘤血管生成之间均有相关性,且Girdin与VEGF表达正相关,提示Girdin与VEGF在胶质瘤的血管生成过程中,促进胶质瘤新生血管的形成,引起胶质瘤细胞增殖、浸润、侵袭及转移,促进胶质瘤的发生发展。Girdin表达与胶质瘤的生物学行为相关,可成为胶质瘤预后的预测因子。

Abstract: Objective:To study the expressions of Girdin and VEGF in glioma and their correlations with the angiogenesis of glioma,and to discuss the role of Girdin and VEGF in glioma angiogenesis. Methods:Using Envision immunohistochemical staining to detect the expressions of Girdin protein and VEGF in normal brain tissue and different grade glioma tissues,and using tumor endothelial cells labeled with CD34 antibody,counting microvascular density(MVD) at the same time,in order to analyze the relationships among Girdin,VEGF and MVD. Results: Girdin and VEGF were both expressed in glioma.But in normal brain tissue,their expressions were not found.Furthermore,the expressions of Girdin and VEGF in high grade gliomas were both significantly higher than those in low grade gliomas.The differences between them were all statistically significant(P<0.05). With the grading of glioma elevated,MVD increased. Statistial difference of MVD were found between high and low grade gliomas(P<0.05). In gliomas,the expression of Girdin was positively correlated with MVD (P<0.01),and the positive correlations were also found between VEGF and MVD(P<0.01), Girdin and VEGF(P<0.01). In gliomas,the expression of Girdin significantly correlated with KPS score, resection range and glioma grade(P<0.05), but not correlated with age and sex. Conclusion: High expressions of Girdin and VEGF can be found in glioma, furthmore with correlations with the angiogenesis of glioma, suggesting that Girdin and VEGF can jointly promote the formation of new blood vessels,thus promoting the proliferation,infiltration,invasion and mestastasis of glioma cells as well as the occurrence and development of glioma. The expression of Girdin is closely related to the biological behavior of glioma. It may be an prognostic factor in glioma.
