尿素维E乳膏中抑菌剂用量研究

Study on the Dosage of Antibacterial Agents in Urea and Vitamin E Cream

摘要:

目的:评估不同生产厂家尿素维E乳膏中抑菌剂的添加量和抑菌效力。方法:采用高效液相色谱法测定尿素维E乳膏中的抑菌剂,色谱柱为Capcell PAK C18250 mm × 4.6 mm5 μm),流动相为甲醇-1%冰醋酸(6040),流速为1 mL·min-1,检测波长为254 nm,进样量为10 μL。按《中华人民共和国药典》进行抑菌效力试验,制备金黄色葡萄球菌、铜绿假单胞菌、大肠埃希菌、白色念珠菌、黑曲霉菌液,添加至10个生产厂家的样品,使样品含菌量约为105106 cfu·g-1,混匀后置2025℃避光贮存,分别在271428 d共计4个时间节点,测定存活菌数。结果:羟苯甲酯、羟苯乙酯、羟苯丙酯的线性范围分别为0.010.1 mg·mL-1,精密度、重复性、稳定性的RSD均小于1%9个生产厂家的样品使用羟苯乙酯作为抑菌剂,浓度为0.05%0.20%1个生产厂家的样品使用羟苯甲酯和羟苯丙酯作为抑菌剂,浓度分别为0.1%0.05%。抑菌效力测定结果显示,细菌2 d下降值的对数大于27 d下降值的对数大于328 d菌数未增加;真菌14 d菌数下降值的对数大于等于228 d菌数未增加,各个生产厂家产品的抑菌效力均符合要求。结论:尿素维E乳膏制剂的抑菌效力较好,抑菌剂添加量差异较大。

Abstract:

Objective: To evaluate the amount and efficacy of preservatives added in urea and vitamin E cream from different manufacturers.Methods: High-performance liquid chromatography HPLC was used to determine the preservatives in urea and vitamin E cream. The chromatographic column was Capcell PAK C18 250 mm × 4.6 mm 5 μm), the mobile phase was methanol-1% glacial acetic acid 6040), the flow rate was 1 mL·min-1 the detection wavelength was 254 nm and the injection volume was 10 μL. The antimicrobial efficacy test was conducted according to the "Pharmacopoeia of the People's Republic of China". Staphylococcus aureus Pseudomonas aeruginosa Escherichia coli Candida albicans and Aspergillus niger suspensions were prepared and added to samples from 10 manufacturers with the bacterial content of the samples being approximately 10⁵ ~ 10⁶ cfu·g⁻¹. After mixing the samples were stored in the dark at 20 ~ 25 ℃. The viable bacterial counts were measured at four time points 2 7 14 28 d.Results: The linear ranges for methylparaben ethylparaben and butylparaben were 0.01 ~ 0.1 mg·mL-1 respectively. The relative standard deviation RSD for precision repeatability and stability is all less than 1%. In the samples from 9 manufacturers ethylparaben was used as the preservative with concentrations ranging from 0.05% to 0.20%. In the sample from 1 manufacturer methylparaben and butylparaben were used as preservatives with concentrations of 0.1% and 0.05% respectively. In the antimicrobial efficacy test the logarithmic reduction of bacterial counts after 2 d was greater than 2 and after 7 d it was greater than 3 no increase in bacterial counts was observed over 28 d. For fungi a logarithmic reduction greater than 2 or equal to in colony counts was observed after 14 d with no increase in counts over 28 d. The antimicrobial efficacy of products from all manufacturers met the requirements.Conclusion: The antibacterial efficacy of the urea and vitamin E cream preparation is good with a significant variation in the amount of antibacterial agent added.
