女贞子水溶性多糖的含量测定

摘要: 目的:建立女贞子中水溶性多糖含量测定的方法。方法:采用蒽酮-硫酸比色法、紫外分光光度法,用供试品空白溶液作为参比、以葡萄糖溶液作为对照品进行测定。结果:在570 nm的波长下,葡萄糖对照品溶液0~0.1 mg·mL-1范围以内,吸光度与浓度的线性关系优良;以葡萄糖(0.1 mg·mL-1)作为对照品时,其线性回归直线方程为A =0.992 3C+0.004 2(r=0.999 2,n=6);女贞子供试品水溶性多糖的含量为8.03 mg·g-1,平均回收率为100.54%,RSD=2.80%(n=6),精密度良好;用蒽酮-硫酸比色法测定女贞子水溶性多糖的含量,以提取女贞子水溶性多糖和葡萄糖对照品经由紫外全波长扫描确定在570 nm处有最大吸光度值,同时进行了线性关系、稳定性、精密度以及回收率等方法学考察,各项指标表现良好。结论:用蒽酮-硫酸法和紫外分光光度法来测定女贞子水溶性多糖的含量,方法简单,操作性强,重复性好。

Abstract: Objective:To establish a method on the content determination of water-soluble polysaccharide in fructus ligustri lucidi. Methods: Both anthranone-sulfuric acid colorimetry and UV visible spectrophotometry were used, which the blank solution of the sample was used as reference and glucose solution as the control sample. Results: Under the wavelength of 570 nm, when glucose reference substance was 0-0.1 mg·mL-1, the linear relationship between the absorbance and the concentration was good. When glucose (0.1 mg·mL-1) as the reference substance, the linear regression line equation was A=0.992 3C+0.004 2(r=0.999 2, n=6). The content of water-soluble polysaccharide was 8.03 mg·g-1, the average recovery was 100.54%, RSD was 2.80%(n=6), and the precision was good. Using anthranone-sulfuric acid colorimetry to determine the content of water-soluble polysaccharide in fructus ligustri lucidi, fructus ligustri lucidi water-soluble polysaccharide and glucose reference substance determined by ultraviolet wavelength scanning all have maximum absorbance at 570 nm, and the methodological indicators such as linearity, stability, precision and recovery were performing well. Conclusion: The method is simple, feasible, and suitable for determining the content of water-soluble polysaccharide in fructus ligustri lucidi.
