X射线辐照处理对光果甘草质量的影响研究

摘要: 目的:研究X射线辐照处理对光果甘草中三萜类及黄酮类有效成分含量及产量的影响。方法:采用辐照剂量分别为5、10、30、50 Gy的X射线辐照处理光果甘草种子,空白对照组为未经X射线辐照处理的光果甘草种子。栽培1年后,采用高效液相色谱(HPLC)法测定各甘草样品中甘草酸、异甘草酸、甘草苷和异甘草苷的含量,并计算其产量,运用SAS分析软件进行统计学分析。结果:随着辐照剂量的增加,各组光果甘草样品的干重、有效成分含量及产量均呈现出先升高后降低再升高的趋势;干重以及三萜类化合物的含量和产量在辐射剂量为5 Gy时达到最大值,黄酮类化合物的含量和产量在辐照剂量为50 Gy时达到最大值。结论:光果甘草经X射线辐照处理后,其黄酮类及三萜类有效成分的含量和产量均明显增加,辐照处理极有可能导致了三萜及黄酮代谢途径功能基因的变异,本研究可为光果甘草的品种改良奠定基础。

Abstract: Objective: To investigate the effects of X-ray irradiation treatment on the content and yield of triterpenes and flavonoids effective component in Glycyrrhiza glabra(G. glabra). Methods:G. glabra seeds were irradiated by X-ray with four different doses, 5, 10, 30, 50 Gy. Seeds without irradiation were in the control group.After cultivating for one year, the content of 18β-glycyrrhizic acid,18α-glycyrrhizic acid, liquiritin, and isoliquiritin in G. glabra were assayed by high performance liquid chromatography(HPLC) and the yield was calculated, and SAS was used to process statistical analysis. Results: With the increase of irradiation dose, the dry weight, effective component content and yield of G. glabra all showed an increase-decrease-increase tendency. The dry weight and content of triterpenes reached the peak at the dose of 5 Gy, while the content and yield of flavonoids reached the peak at the dose of 50 Gy. Conclusion:X-ray irradiation significantly increases the content and yield of triterpenes and flavonoids in G. glabra. The reason is speculated that variations of function genes in triterpenes and flavonoids metabolic pathways can be induced by irradiation treatment. This study will lay the foundation for the breed improvement of G. glabra.
