Rapid and Simultaneous Determination of Seven Kinds of Neurotransmitters in Rat Plasma by HPLC-MS/MS

摘要: 目的:建立高效液相色谱串联质谱(HPLC-MS/MS)同时快速检测大鼠血浆中多巴胺(DA)、去甲肾上腺素(NE)、肾上腺素(E)、5-羟色胺(5-HT)、谷氨酸(Glu)、γ-氨基丁酸(GABA)、乙酰胆碱(Ach)7种神经递质含量的方法。方法:采用Waters XSelect@HSS PFP色谱柱(氟苯基柱,2.1 mm×150 mm, 3.5 μm),以0.1%甲酸水和0.1%甲酸乙腈为流动相,梯度洗脱。在全扫描和MS2扫描下,比较正、负离子模式,确定定性离子对和定量离子对。结果:7种神经递质在4 min内全部出峰,在1~500 μg·L-1范围内线性关系良好,最低检出限(S/N=3)为0.1~0.5 μg·L-1。结论:该方法操作简便、灵敏度高、结果准确,可用于快速检测大鼠血浆中7种神经递质的含量。

Abstract: Objective:To established a rapid high performance liquid chromatography mass spectrometry method for simultaneous determination of dopamine(DA),norepinephrine(NE), epinephrine(E), serotonin(5-HT), glutamate(Glu), γ-aminobutyric acid(GABA) and acetylcholine (Ach) . Methods: A Waters XSelect@HSS PFP column(2.1 mm×150 mm, 3.5 μm) was used for the sample separation with 0.1% formic acid water and 0.1% formic acid acetonitrile as the mobile phase under gradient elution. By the full scanning and MS2 scanning, the positive and negative ion modes were compared, and the qualitative ion pair and quantitative ion pair were determined. Results:7 kinds of neurotransmitters were determined in 4 min, the calibration curves showed good linearity within the concentrations of 1~500 μg·L-1, and the limits of detection were 0.1 to 0.5 μg·L-1. Conclusion: The method is simple,sensitive and accurate. It is suitable for rapid determination of the 7 kinds of neurotransmitters in rat plasma.
