摘要

目的:通过对人羊水干细胞(hAFSCs)分化过程中表观遗传调控机制研究的相关文献进行可视化分析,分析该领域国内外的研究热点及未来趋势,旨在深化对 hAFSCs 多能性维持与分化分子机制的理解,并为细胞治疗、再生医学以及新型治疗策略的开发提供理论基础。方法:采用主题词检索法,以“人羊水干细胞”“分化”“表观遗传”及“甲基化”为主题词,在PubMed数据库中检索20011月至20245月的相关文献。利用Excel软件绘制年度发文量趋势图,运用CiteSpace 6.2.R4工具绘制知识图谱,对作者、国家、关键词等节点进行分析。结果:本研究共纳入601篇文献,这些文献涉及全球987位研究者Collas Philippe是该领域最为活跃的作者之一,发表的文章最多(number=14)。关键词共现分析显示,“再生医学”与“基因表达”是该领域近年来研究的重点。关键词突现分析揭示了“表观遗传修饰”“耐药”“诱导多能干细胞”“干细胞研究”及“DNA羟甲基化”等为当前研究热点,这些热点为后续研究提供了方向。结论:近年来,hAFSCs分化过程中的表观遗传调控机制研究取得了显著进展,不仅揭示了更多关键的调控因子和信号通路,还深化了对细胞命运决定机制的理解。未来应聚焦热点,深挖hAFSCs分化机制,关注表观遗传精细调控,以推动精准细胞治疗策略,促进细胞治疗与再生医学的创新发展。

Abstract

Objective: Through visual analysis of the research literature on epigenetic regulatory mechanisms during the differentiation of human amniotic fluid stem cells hAFSCs), to analyze the research hotspots and future trends in this field at home and abroad aiming to deepen the understanding of the molecular mechanisms of pluripotency maintenance and differentiation of hAFSCs and provide a solid theoretical basis for cell therapy regenerative medicine and the development of novel treatment strategies.Methods: With "human amniotic fluid stem cells" "differentiation" "epigenetic inheritance" and "methylation" as the main topics the relevant literatures from January 2001 to May 2024 were searched in PubMed database. Excel software was used to draw the trend chart of the annual publication volume and CiteSpace 6.2.R4 tool was used to draw the knowledge graph and nodes such as author country and keywords were analyzed.Results: The study included 601 articles contributed by 987 researchers worldwide with Collas Philippe being one of the most active authors in this field with the most published articles number=14. Keyword co-occurrence analysis shows that "regenerative medicine" and "gene expression" are the focus of research in this field in recent years. Keyword breakout analysis revealed that "epigenetic modification" "drug resistance" "induced pluripotent stem cells" "stem cell research" and "DNA hydroxymethylation" were the current research hotspots which provided the direction for subsequent research.Conclusion: In recent years significant progress has been made in epigenetic regulatory mechanisms during hAFSCs differentiation which not only reveals more key regulatory factors and signaling pathwaysbut also deepens the understanding of cell fateful mechanisms. The future researches we should focus on hot spots dig deep into the differentiation mechanism of hAFSCs and pay attention to the fine epigenetic regulation so as to promote the precision cell therapy strategy and promote the innovative development of cell therapy and regenerative medicine.
