基于周细胞PDGFRβ信号研究通脑饮“祛瘀生新”促进缺血性脑卒中后血管新生的作用机制研究(MS2022016)

摘要

目的:为进一步科学、完备地研究祛瘀生新方作用机制,传承国医大师周仲瑛教授临床经验,使用网络药理学和分子对接技术,探究其治疗脑梗死的作用机制。方法:通过TCMSP数据库、HERB数据库和文献检索获取祛瘀生新方的中药有效活性成分以及作用靶点,同时分析以协定方祛瘀生新方治疗的典型病例。结果:临床使用证实祛瘀生新方可有效改善脑梗死患者神经功能缺损症状,改善患者预后。本研究显示,祛瘀生新方存在96个有效成分,合并479个作用靶点,GO富集分析包含4046个条目,KEGG通路富集分析包含199条KEGG通路,分子对接结果显示关键成分和核心靶点具有较好的结合作用。祛瘀生新方的主要活性成分为花生四烯酸、辣椒素、山柰酚、苯丙二酸、钩藤碱,可能通过干预脂质与动脉粥样硬化、鞘脂信号通路等,激活脑-肾-肠轴发挥治疗脑梗死的作用。结论:传承协定祛瘀生新方通过祛脑瘀、生新脉、行血运治法,治疗脑梗死,改善患者预后,其潜在的作用机制可能与血管生成、细胞凋亡、抗炎、脑-肾-肠轴等生物进程密切相关,为后续的实验研究、新药开发提供了线索和依据。

Abstract

Objective: To conduct a more scientific and comprehensive study on the action mechanism of quyu shengxin formula and inherit the clinical experience of professor Zhou Zhongying, network pharmacology and molecular docking techniques were employed to explore its mechanism of action in treating cerebral infarction.Methods: The effective active components and target proteins of the quyu shengxin formula were obtained through the TCMSP database, HERB database, and literature retrieval. Meanwhile, typical patient cases treated with the agreed prescription quyu shengxin formula were analyzed.Results: The clinical application of quyu shengxin formula has confirmed that it can effectively alleviate the neurological deficit symptoms of cerebral infarction patients and improve their prognosis. Network pharmacology research revealed that the quyu shengxin formula consists of 96 effctive components and 479 target proteins. The GO enrichment analysis encompassed 4046 items. The KEGG pathway enrichment analysis included 199 KEGG pathways. The molecular docking results indicated that the key components and core target proteins exhibited favorable binding effects. The main active components of the quyu shengxin formula are arachidonic acid, capsaicin, kaempferol, phenylmalonic acid, and rhynchophylline. It may exert the therapeutic effect on cerebral infarction by interfering in lipid and atherosclerosis, sphingolipid signaling pathways, and activating the brain-kidney-intestine axis.Conclusion: Traditional agreed prescription quyu shengxin formula, which aims to remove brain stasis, generate new vessels, and promote blood circulation for treating cerebral infarction, can improve the prognosis of patients. Its potential mechanism of action may be closely related to biological processes such as angiogenesis, apoptosis, anti-inflammation, and the brain-kidney-intestine axis, providing clues and a basis for subsequent experimental research and new drug development.
