黄芩汤颗粒剂中黄芩苷、甘草酸及芍药苷的含量测定研究

Content Determination of Baicalin,Glycyrrhizic Acid and Paeoniflorin in Huangqin Decoction Granule

摘要: 目的:测定黄芩汤颗粒剂主要成分的含量。方法:采用高效液相色谱法测定处方黄芩中的黄芩苷,甘草中的甘草酸和白芍中的芍药苷含量,以Kromasil C18(4.6 mm×250 mm,5 μm)色谱柱,应用 0.05%磷酸和乙腈梯度洗脱并进行方法学考察。结果:高效液相色谱法测出在黄芩苷3.6~57.60 μg·mL-1,r=0.9996;甘草酸在4.16~52 μg·mL-1,r=0.9999;芍药苷在2.85~22.8 μg·mL-1,r=0.9999 范围内呈良好的线性关系,黄芩苷、甘草酸及芍药苷的平均回收率分别为 96.31%,97.69%和97.27%;RSD分别为1.29%,1.03% 和1.29%。结论:建立的含量测定方法简便、重现性好,可用于黄芩汤颗粒剂的质量控制。

Abstract: Objective: To establish a method of content determination on baicalin, glycyrrhizic and paeoniflorin of huangqin decoction granule.Methods: The content of baicalin,glycyrrhizin and paeoniflorin was measured by HPLC, the column was Kromasil C18 (4.6 mm×250 mm, 5 μm), the mobile phase was selected 0.05% phosphoric acid and acetonitrile with a gradient elution. Results:The linear ranges of baicalin,glycyrrhizin and paeoniflorin were within 3.6~57.60 μg·mL-1, r=0.9996; 4.16~52 μg·mL-1, r=0.9999; 2.85~22.8 μg·mL-1, r=0.9999,and the average recoveries were 96.31% (RSD of 1.29 %), 97.69% (RSD of 1.03 %) and 97.27% (RSD of 1.29 %). Conclusion: The method is simple and reproducible, and can be used for the quality control of Huangqin decoction granule.
