Quantitative Determination of Free Gallic Acid and Total Gallic Acid in Euphorbiae Humifusae Herba by HPLC

摘要: 目的:建立HPLC法测定地锦草中游离没食子酸和总没食子酸的含量。方法:采用Agela Odyssil C18(4.6 mm×250 mm,5 μm)色谱柱;流动相为甲醇-0.5%磷酸溶液(8∶92);检测波长270 nm;流速1.0 mL·min-1;柱温30 ℃。结果:没食子酸在0.034 0~1.700 2 μg范围内呈良好线性关系(r=1.000 0),游离没食子酸和总没食子酸平均回收率分别为99.63%和99.34%。结论:该方法操作简便准确,灵敏度高,重复性好,可用于地锦草中没食子酸的质量控制。

Abstract: Objective:To establish a method for quantitative determination of free gallic acid and total gallic acid in Euphorbiae Humifusae Herba by HPLC.Methods:The HPLC procedure was performed on the chromatographic column of Agela Odyssil C18(4.6 mm×250 mm, 5μm)at 30℃, the mobile phase was methanol-0.5% phosphoric acid solution(8∶92)with the flow velocity of 1.0 mL·min-1, and the detection wavelength was 270 nm.Results: Gallic acid showed a good linear relationship with peak area at a range from 0.0340 μg to 1.7002 μg(r= 1.0000), the free gallic acid recovery was 99.63% and the total gallic acid recovery was 99.34%.Conclusion:The method is simple, accurate with high sensitivity and good repeatability, and can be used for the quality control of gallic acid in Euphorbiae Humifusae Herba.
