Antioxidant and Anti-inflammatory Properties of the Citrus Flavonoids Hesperidin and Hesperetin: An Review of Their Molecular Mechanisms

摘要: 炎症和氧化应激是威胁生命健康的两个主要危险因素。橙皮苷(hesperidin, Hsd)及其糖苷配基橙皮素(hesperetin, Hst)是来自芸香科柑橘属植物果实的两种黄酮类化合物,具有抗氧化和抗炎等多种生物学活性。Hsd/Hst的抗氧化活性不仅限于自由基清除,而且还能通过ERK/Nrf2信号通路增强抗氧化细胞防御功能。多个体外和体内研究评估了Hsd,其代谢物或其合成衍生物可通过抑制相关炎症信号通路的激活,减少炎性靶点(NF-κB,iNOS,COX-2)以及慢性炎症标志物,起到抗炎效果。本文就近些年有关Hsd和Hst的抗氧化和抗炎作用及其相关分子机制的研究进行综述。

Abstract: Inflammation and oxidative stress are two major causes of various life-threatening diseases. Hesperidin (Hsd) and its aglycone, hesperetin (Hst), are two flavonoids from citrus species that have numerous biological properties, particularly antioxidant and anti-inflammatory. New findings showed that the antioxidant activity of Hsd/Hst was not only limited to its radical scavenging activity, but it augmented the antioxidant cellular defenses via the ERK/Nrf2 signaling pathway as well. Various in vitro and in vivo studies have been conducted to evaluate Hsd, its metabolites, or its synthetic derivatives which could play an anti-inflammatory role by inhibiting the activation of relevant inflammatory signaling pathways, reducing inflammatory targets including NF-κB, iNOS, and COX-2, and the markers of chronic inflammation. In this review, new findings regarding the antioxidant and anti-inflammatory effects of Hsd and Hst and the related molecular mechanisms are discussed.
