UPLC-MS/MS法同时检测参芪健胃颗粒中芍药苷、橙皮苷、甘草酸铵的含量

Determination of Paeoniflorin, Hesperidin, Ammonium glycyrrhizinate in Shenqi Jianwei Granules by UPLC-MS/MS

摘要: 目的:建立一种超高效液相-串联质谱(UPLC-MS/MS)方法同时测定参芪健胃颗粒中芍药苷、橙皮苷、甘草酸铵的含量。方法:色谱柱为Agilent Eclipse Plus C18型 (2.1 mm×50 mm,1.8 μm),流动相为乙腈-0.1%甲酸水溶液,梯度洗脱,流速为0.3 mL·min-1,柱温为35 ℃。采用负离子多反应监测(MRM)模式同时测定提取物中芍药苷、橙皮苷、甘草酸铵的含量,并以氯霉素为内标。结果:芍药苷、橙皮苷、甘草酸铵的线性范围分别为0.044~11.24、0.021~10.924、0.016~5.275 μg·mL-1(r>0.999 0);定量限分别为11.0、7.0、8.0 ng·mL-1;检测限分别为4.4、3.5、4.0 ng·mL-1;精密度、重复性、稳定性良好;平均加样回收率95%~105%(RSD<6.5%,n=6)。结论:该方法简单快捷、灵敏度高、专属性强,可同时检测参芪健胃颗粒中芍药苷、橙皮苷、甘草酸铵的含量。

Abstract: Objective: To establish an ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method for determination of paeoniflorin, hesperdidin, ammonium glycyrrhizinate in shenqi jianwei granules. Methods:The determination was performed on Agilent Eclipse Plus C18 column (2.1 mm×50 mm,1.8 μm), with the mobile phase consisted of acetonile-0.1% formic acid (gradient elution) at the fluid speed of 0.3 mL·min-1. The column temperature was set at 35 ℃. Multiple reaction monitoring (MRM) in negative mode was used for the quantification of paeoniflorin, hesperidin, ammonium glycyrrhizinate, using chloramphenicol as an internal standard (IS). Results:The liner range of paeoniflorin, hesperidin, ammonium glycyrrhizinate were 0.044-11.24, 0.021-10.924, 0.016-5.275 μg·mL-1, respectively (r>0.999 0). The limits of quantitation were 11.0, 7.0, 8.0 ng·mL-1. The limits of detection were 4.4, 3.5, 4.0 ng·mL-1 respectively. The results demonstrated satisfying precision, repeatability, and stability. Average recoveries were 95%~105% (RSD<6.5%, n=6). Conclusion:The established UPLC-MS/MS method is accurate, simple, sensitive and specific, which can be used for determination of paeoniflorin, hesperidin, ammonium glycyrrhizinate in shenqi jianwei granules.
