金属元素检测技术在药包材常规检测、相容性研究中的应用及探讨

摘要: 包材是指药品生产企业生产的药品和医疗机构配制的制剂所使用的直接接触药品的包装材料和容器,分为玻璃、金属、塑料、橡胶、预灌封等类型。药包材常规检测即按照《国家药包材标准》(2015)中规定的检验方法及技术要求,对药包材的质量进行监管。药品的相容性研究则是证明药品与包装材料之间没有发生严重的相互作用,并导致药品稳定性和有效性发生改变,或者产生安全性风险的过程。药包材本身的质量、安全性、使用性能以及药包材与药品之间的相容性对药品质量有着十分重要的影响。药包材中含有的砷、汞、铅、镉等重金属元素不仅可能会引起药效降低,而且进入人体后因无法消化、排除,经长期累积会对人体造成损伤。加强对药包材中金属元素含量的监管,提升金属元素检测水平,不仅是保障人民群众健康用药的客观需要,也是药品一致性评价及关联评审的必然要求。本文从药包材材料类型出发,综述了在药包材中常规检测及相容性研究中金属元素检测技术的应用情况,主要包括原子吸收光谱法、紫外-可见分光光度法、比色法、电感耦合等离子体原子发射光谱法、电感耦合等离子体质谱法等方法,并就应用实践中产生的问题提出解决方案,以期对未来的检测工作提供建设性意见。

Abstract: Pharmaceutical packaging materials refer to the packaging materials and containers that are directly in contact with the pharmaceuticals produced by pharmaceutical manufacturers and the preparations made by medical institutions, which are divided into glass, metal, plastic, rubber, pre-filling and other types. Conventional testing of pharmaceutical packaging materials means to supervise the quality of pharmaceutical packaging materials in accordance with the inspection methods and technical requirements stipulated in the national standards for pharmaceutical packaging materials (2015).The compatibility study of drugs is a process to prove that there is no serious interaction between drugs and packaging materials, which leads to changes in drug stability and effectiveness, or safety risks.The quality, safety, performance and compatibility of pharmaceutical packaging materials have important influence on drug quality.The heavy metal elements such as arsenic, mercury, lead and cadmium contained in the pharmaceutical packaging materials may not only cause the drug effect to decrease, but also cause damage to human body because of indigestibility and less-elimination.Strengthening the supervision on the content of metal elements in pharmaceutical packaging materials and improving the detection level of metal elements are not only the objective needs for ensuring drug safety for the public, but also the inevitable requirements of drug consistency evaluation and joint review. Based on conventional detection and compatibility studies, this research reviews the recent detection technology of metal elements in pharmaceutical packaging materials according to the material type,including atomic absorption spectrometry, uv-vis spectrophotometry, colorimetric method, inductively coupled plasma atomic emission spectrometry and inductively coupled plasma mass spectrometry. Besides, the problems encountered in the actual testing are discussed in detail to provide constructive suggestions for the future testing work.
