基于网络药理学研究黄龙汤治疗便秘的作用机制

Study on the Mechanism of Huanglong Decoction Treating Constipation Based on Network Pharmacology

摘要: 目的:采用网络药理学方法探讨黄龙汤治疗便秘的可能机制。方法:通过检索TCMSP、UniProt、DisGeNET、CTD、TTD等数据库,收集黄龙汤化学成分、靶点及便秘靶点;利用STRING数据库和Cytoscape软件建立PPI网络和黄龙汤成分-靶点-疾病网络;在Metascape平台对核心靶点进行GO生物功能和KEGG通路富集分析。结果:从黄龙汤中共筛选出化学成分182个,对应靶点278个,与便秘相关的靶点203个,其中超过平均度值的关键靶点39个;关键成分有黄酮类、甾醇类等,核心靶点有ESR1、AR、PTGS1等;主要生物过程有节律过程、血液循环等;主要信号通路有 cAMP通路、5-羟色胺能突触、PI3K-Akt通路等。结论:黄龙汤可能是通过多成分调节多靶点、多条信号通路发挥治疗便秘的作用。

Abstract: Objecetive:To Study the potential mechanism of huanglong decoction in treating constipation based on network pharmacology. Methods: The TCMSP database, UniProt datebase, CTD database, TTD database and DisGeNET database were used to screen the chemical components, corresponding targets of huanglong decoction and the targets related to constipation. The drug compound-target-disease network and target protein interaction PPI network were constructed through Cytoscape3.7.1 software and STRING database. Metascape online platform were used for the GO function and KEGG pathway enrichment analysis of the targets. Results: 182 chemical components were obtained in huanglong decoction, and 278 corresponding targets were obtained. 203 common targets were related with constipation, among which 39 key targets exceeded the average degree value. The main components flavonoids and sterols, and ESR1,AR, PTGS1 were the key targets. The main biological processes include rhythm process and blood circulation, and the main signal pathways include cAMP pathway, serotonergic synapse, PI3K-Akt pathway. Conclusion: Huanglong decoction may play the role in the treatment of constipation through multi-component regulation, multi-target and multiple signal pathways.
