自噬参与调控骨改建的研究进展

摘要: 骨改建是成骨细胞、破骨细胞、骨细胞参与的一个紧密联系的动态过程。骨代谢性疾病常引起骨改建失衡,导致骨结构异常。细胞自噬是一种广泛存在于真核生物细胞,且进化上高度保守的细胞降解过程。在不同的细胞微环境下,自噬对细胞保护或破坏是不同的。研究发现自噬在骨改建的过程中发挥重要作用,自噬的缺损可能导致骨细胞功能障碍;自噬及其相关蛋白参与成骨细胞、破骨细胞、骨细胞的分化及功能,从而维持骨改建的稳定。然而,自噬相关信号通路错综复杂。本研究基于当前国内外有关自噬参与骨改建的研究进展,从自噬起源、信号通路以及自噬与骨改建、自噬与骨代谢疾病等方面展开综述,以期为自噬参与骨改建的进一步研究提供理论支持。

Abstract: Bone remodeling is a dynamic process involving in osteoblasts, osteoclasts and osteoblasts. Bone metabolic diseases often lead to imbalance of bone remodeling and abnormal bone structure. Autophagy is a highly conserved cell degradation process that widely exists in eukaryotic cells. Autophagy protects or destroys cells differently in different cell microenvironments. Studies have found that autophagy, which plays an important role in bone remodeling, is involved in the differentiation and function of osteoblasts, osteoclasts and osteoblasts. It can maintain the stability of bone remodeling. Signaling pathways of autophagy are complex. Based on current research progress, this study reviews from the aspects of the origin of autophagy, signal pathway, bone remodeling and bone metabolic diseases, in order to provide theoretical support for further research.
