正交试验优化安肠液的水提及醇沉工艺

Optimization of Water Extraction and Ethanol Precipitation Processes for Anchang Liquid by Orthogonal Design

摘要: 目的:优化安肠液最佳水提及醇沉工艺。方法:采用正交试验以黄芪甲苷含量和干膏率为指标,以浸泡时间、加水量、提取时间、提取次数为考察因素优选安肠液最佳水提工艺;以药液相对密度、药液含醇量、放置时间为考察因素,优选安肠液最佳醇沉工艺。结果:所建立的黄芪甲苷含量方法的线性范围为0.0241~0.3856 g·mL-1(r=0.999 6),精密度、重复性、稳定性及准确度均良好。安肠液最佳水提工艺为加8倍量水浸泡30 min,煎煮时间1.0 h,煎煮3次;最佳醇沉工艺为水提液浓缩至相对密度为1.06 g·mL-1(25℃),药液含醇量为80%,放置时间为12 h。结论:优化的安肠液水提及醇沉工艺稳定、可行。

Abstract: Objective:To optimize the water extraction and ethanol precipitation process of anchang liquid.Methods:The orthogonal text was used to study the four factors including the soak time, amount of water added, extraction time and frequency by taking the content of astragaloside IV and dry extract yield as indicators to optimize the water extraction process of anchang liquid. Taking the relative density of the drug solution, ethanol content, and the storage time as the investigating factors, the best ethanol precipitation process of anchang liquid was selected. Results:The established method for content determination of astragaloside IV with the linearity range of 0.024 1~0.385 6 g·mL-1(r=0.999 6) had good precision, reproducibility, stability and accuracy. The best water extraction process of anchang liquid was to add with 8 times water, soak for 30 min, decoct for 1 hour for three times. The optimal ethanol precipitation process was to concentrate the water extraction to a relative density of 1.06 g·mL-1 (25℃),with 80% ethanol content and 12 h storage time.Conclusion:Optimized water extraction and ethanol precipitation processes for anchang liquid were stable and feasible.
