基于小鼠模型筛选黄芪当归治疗2型糖尿病的最佳配比及最优半仿生提取工艺

Screening of the Optimal Ratio of Astragalus-Angelica and the Optimal Semi-Bionic Extraction Process for Treating Type 2 Diabetic in Mice Model

摘要: 目的:寻找黄芪、当归治疗2型糖尿病小鼠的最佳配比及最优半仿生提取工艺。方法:以2型糖尿病小鼠血糖值、进食量和体重为指标,通过口服灌胃不同比例黄芪当归的水煎液,筛选出当归、黄芪治疗2型糖尿病小鼠的最佳配比;运用中心组合设计实验,以2型糖尿病小鼠第14天和第0天的血糖差值、进食量差值、和体重差值为指标,优化当归、黄芪半仿生提取的最佳工艺条件。结果:当归、黄芪治疗2型糖尿病小鼠的最佳配比是黄芪∶当归为7∶2;半仿生提取当归、黄芪的最佳工艺条件为第1次提取人工胃液用量为7.7倍,第1次提取时间为2.6 h,第2次提取人工肠液用量为6.2倍,第2次提取时间为3.5 h。结论:本实验确定了当归、黄芪治疗2型糖尿病小鼠的最佳配比及半仿生提取的最佳工艺条件,为临床上使用当归、黄芪治疗2型糖尿病提供了一定依据。

Abstract: Objective: To explore the optimal ratio and the optimal semi-bionic extraction process for astragalus and angelica treating type 2 diabetic mice. Methods: Taking the blood glucose level, food intake and body weight of mice with type II diabetic as indicators, the optimal ratio of astragalus and angelica in the treatment of type 2 diabetes was selected by orally gavage with different proportions of astragalus and angelica decoction. Utilizing the central combination design experiments to optimize the process of semi-bionic extraction of astragalus and angelica with the differences in blood sugar lowering, food intake, and body weight on the 14th day and the 0th day of treatment in type II diabetic mice.Results: The optimal proportion of astragalus and angelica in the treatment of type II diabetic mice was 7∶2. The optimal process for semi-bionic extraction of angelica and astragalus were that the amount of artificial gastric juice for the first time extraction was 7.7 times, extraction duration was 2.6 h, and the amount of artificial intestinal fluid used for the second extraction was 6.2 times, and the second extraction duration was 3.5 h.Conclusion: The results of this experiment provide a basis for the clinical use of angelica and astragalus in the treatment of type II diabetes.
