贞芪扶正颗粒治疗乳腺癌的网络药理学研究

Network Pharmacology Study of Zhenqi Fuzheng Granule in the Treatment of Breast Cancer

摘要: 目的:基于网络药理学探讨贞芪扶正颗粒治疗乳腺癌的潜在机制,以期为乳腺癌的治疗提供更加可靠的理论基础。方法:通过检索TCMSP数据库获取贞芪扶正颗粒的有效成分和作用靶点,并检索GEO数据库得到乳腺癌相关靶点,使用R语言绘制乳腺癌差异基因表达热图,进行基因本体(GO)富集分析和KEGG通路富集分析,通过STRING数据库构建PPI网络,Cytoscape 3.7.2 构建有效成分-靶点网络图及靶点-KEGG通路网络图。结果:共得到有效成分31种,对应靶点598个,其中有效成分与乳腺癌共同靶点34个,GO富集分析得到生物过程(BP)条目495个,细胞组分(CC)条目6个,分子功能(MF)条目42个,BP富集显著的条目主要包括对机械刺激的反应、脂多糖应答、对细菌来源分子的反应、化学应激的细胞反应等。KEGG通路富集分析共得到47条通路,其中乙型肝炎、卡波西肉瘤相关疱疹病毒感染、IL-17信号通路、细胞衰老、爱泼斯坦-巴尔病毒感染、人T细胞白血病病毒1感染等通路富集显著。结论:贞芪扶正颗粒主要通过有效成分如木犀草素、异黄酮、芒柄花黄素、山柰酚、槲皮素等作用于JUN、E2F1、FOS、E2F2、MYC和STAT1等靶点来调控乙型肝炎、卡波西肉瘤相关疱疹病毒感染、IL-17信号通路、细胞衰老、爱泼斯坦-巴尔病毒感染、人T细胞白血病病毒1感染等通路,从而发挥治疗乳腺癌的作用。

Abstract: Objective: To explore the potential mechanism of zhenqi fuzheng granule in the treatment of breast cancer based on network pharmacology, and to provide a more reliable theoretical basis for the treatment of breast cancer. Methods: The effective components and targets of zhenqi fuzheng granule were obtained by searching TCMSP, and the related targets of breast cancer were obtained by searching GEO database. R software was used to draw the differential gene expression heat map of breast cancer, GO enrichment analysis and KEGG pathway enrichment analysis. PPI network is constructed by STRING database, and effective component-target network and target-KEGG path network are constructed by Cytoscape 3.7.2. Results: There are 31 effective components, 598 corresponding targets, and 34 are the intersection targets of effective components and breast cancer. There are 495 BP entries, 6 CC entries and 42 MF entries in GO enrichment analysis. The significant BP enrichment entries mainly include the response to mechanical stimulus,response to lipopolysaccharide,response to molecule of bacterial origin,cellular response to chemical stress. A total of 47 pathways were identified by KEGG pathway enrichment analysis, among which Hepatitis B,Kaposi sarcoma associated herpesvirus infection,IL-17 signaling pathway, Cellular senescence,Epstein Barr virus infection,Human T cell leukemia virus 1 infection were significantly enriched. Conclusion: Zhenqi fuzheng granules mainly act on targets included JUN, E2F1, FOS, E2F2, MYC, STAT1 through its effective components like luteolin, Isoflavanone, formononetin, kaempferol and quercetin, and to regulate pathways such as Hepatitis B,Kaposi sarcoma associated herpesvirus infection,IL-17 signaling pathway,Cellular senescence,Epstein Barr virus infection,Human T cell leukemia virus 1 infection to play a role in the treatment of breast cancer.
