姜紫丹乳膏的制备工艺及其对银屑病小鼠模型的药效学研究

摘要: 目的: 通过星点设计多因素考察对姜紫丹乳膏的处方工艺进行筛选,并探讨其对小鼠尾根部皮肤银屑病模型的治疗作用。方法: 以乳膏的光泽度、均匀度、泛白吸收度和稳定性为评价指标,采用星点设计对影响乳膏质量较大的处方条件进行优化,并对比空白对照组、阳性组、姜紫丹凡士林组和姜紫丹乳膏组对小鼠尾根部皮肤的影响。各组乳膏均匀涂抹至约小鼠尾一半,每日1次,连续给药14 d后,HE染色,观察各组别小鼠尾鳞片表皮颗粒层形成的差异。结果: 姜紫丹乳膏最佳处方为,硬脂酸1.5 g,薄荷油2.05 g,冰片0.17 g。HE染色结果表明,与空白对照组相比,姜紫丹乳膏组小鼠尾根部鳞片表皮颗粒层增多(P<0.01);姜紫丹乳膏组小鼠尾根部鳞片表皮颗粒层形成与姜紫丹凡士林组差异有统计学意义(P<0.05)。结论: 该处方工艺条件下制备的姜紫丹乳膏稳定、光泽、均匀且易吸收,并且对小鼠尾根部鳞片表皮颗粒层形成有明显的促进作用,为临床上使用姜紫丹处方治疗银屑病提供参考依据。

Abstract: Objective: To screen the preparation process of Jiangzidan cream by central composite design multi-factor investigation, and to explore its efficacy on mouse tail root skin psoriasis model. Methods: The gloss, uniformity, whitening absorption and stability of the cream were used as evaluation indicators, and the central composite design was used to optimize the prescription conditions that affect the quality of the cream. The blank control group, the positive group, and the Jiangzidan vaseline group were compared with the Jiangzidan cream group. In each group, the cream was evenly applied to about half of the tail of the mouse once a day. After continuous dosing 14 d, the difference in the formation of the granular layer of the tail scales of the mice in each group was observed after HE staining. Results: The best prescription of Jiangzidan cream is: stearic acid 1.5 g, peppermint oil 2.05 g, borneol 0.17 g. The results of HE staining showed that compared with the blank control group, Jiangzidan cream can promote the formation of the granular layer of the tail scales of the mice(P<0.01), and the formation of the granular layer of the tail scales of the mice in Jiangzidan cream group had statistical difference with that in Jiangzidan vaseline group(P<0.05). Conclusion: The Jiangzidan cream prepared under the established preparation process is stable, glossy, uniform and easy to absorb, and it can obviously promote the formation of the granular layer of the tail scales of the mice, which provide reference for the clinical treatment of psoriasis.
