miR-184对胃癌细胞增殖和迁徙的作用研究

The Effect of miR-184 on the Proliferation and Migration of Gastric Cancer Cells

摘要: 目的: 探究miR-184对胃癌细胞增殖和迁徙的作用及其机制。方法:利用real-time PCR技术检测人胃癌组织及癌旁组织中miR-184的含量,继而检测正常胃黏膜细胞及多种胃癌细胞系中miR-184的含量;利用miR-184抑制剂(miR-184抑制剂组)和miR-184阴性对照序列(对照组)分别转染胃癌细胞系MNK-45后,利用CCK-8法检测该细胞系增殖能力变化;利用划痕实验和Transwell实验检测细胞的迁徙能力;利用Western Blot技术检测细胞Ki67、上皮性钙黏附蛋白(E-cadherin)、神经性钙黏附蛋白(N-cadherin)表达水平。结果:与癌旁组织相比,胃癌组织中miR-184高表达(P<0.05);与正常胃黏膜细胞相比,多种胃癌细胞系中miR-184高表达(P<0.05);与对照组相比,miR-184抑制剂能够显著下调胃癌细胞系MNK-45内miR-184水平(P<0.05),并抑制MNK-45细胞增殖和迁徙(P<0.05);与对照组相比,miR-184抑制剂降低了Ki67和N-cadherin的表达(P<0.05),提高了E-cadherin的表达(P<0.05)。结论:胃癌组织及胃癌细胞系内高表达miR-184,下调miR-184可抑制胃癌细胞的增殖和迁徙。

Abstract: Objective: To explore the effect of miR-184 on the proliferation and migration of gastric cancer cells. Methods: Real-time PCR was used to detect the miR-184 level in human gastric cancer tissues and adjacent tissues, as well as in normal gastric mucosa cells and various gastric cancer cell lines. Gastric cancer cell line MNK-45 were transfected with miR-184 inhibitor (inhibitor group) and miR-184 negative control sequence (control group). CCK-8 assay was used to detect the cell proliferation. Scratch test and Transwell test were used to detect cell migration. The protein levels of Ki67, E-cadherin and N-cadherin were detected by Western Blot. Results: The level of miR-184 in gastric cancer tissues was higher than that in adjacent tissues (P<0.05). Compared with normal gastric mucosa cells, miR-184 was highly expressed in many gastric cancer cell lines (P<0.05). Compared with the control group, miR-184 inhibitor significantly decreased the level of miR-184 in gastric cancer cell line MNK-45 (P<0.05). Compared with the control group, miR-184 inhibitor suppressed the proliferation and migration of MNK-45 (P<0.05). Moreover, miR-184 inhibitor decreased the expression of Ki67 and N-cadherin(P<0.05), but increased the expression of E-cadherin(P<0.05). Conclusion: miR-184 is highly expressed in gastric cancer tissues and gastric cancer cell lines. Down regulation of miR-184 may inhibit the proliferation and migration of gastric cancer cells.
