HPLC法测定制何首乌饮片中2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷含量的不确定度评估

摘要: 目的:对HPLC法测定制何首乌饮片中2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷含量的不确定度进行评估,以期为全面评价分析方法的准确性提供参考。方法:通过建立2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷含量测定的数学模型,分析影响不确定度的因素,并对每个因素进行评价。色谱柱:Agilent ZORBAX SB-C18色谱柱(4.6 mm×250 mm,5 μm);流动相:乙腈-水(25∶75);流速:1.0 mL·min-1;进样体积10 μL;柱温:30 ℃;检测波长:320 nm。结果: 2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷重复性试验的RSD=0.76%(n=6),平均回收率为99.2%(RSD=0.84%,n=9)。置信概率P为95%时,制何首乌饮片中2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的扩展不确定度为0.02%。结论:所建立的不确定度分析方法可用于制何首乌饮片中2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷含量的不确定度评估。

Abstract: Objective: To evaluate the uncertainty of 2, 3, 5, 4′-tetrahydroxystilbene-2-O-β-D-glucos ide in processed polygonum multiflorum by HPLC, in order to provide reference for comprehensive evaluation of the accuracy of analytical methods. Methods: By establishing a mathematical model for the determination of 2, 3, 5, 4′-tetrahydroxystilbene-2-O-β-D-glucos ide.The factors of influencing the uncertainty was analyzed and evaluated for each factor.The Agilent ZORBAX SB-C18(4.6 mm×250 mm,5 μm)was used.The mobile phase consisted of acetonitrile and purified water was 25∶75 and at a flow rate of 1.0 mL·min-1. The injection volume of was 10 μL. The column temperature was set at 30 ℃ and detection wavelength was 320 nm. Results: The RSD of the repeatability test of 2, 3, 5, 4′-tetrahydroxystilbene-2-O-β-D-glucos ide was 0.76% (n=6), the average recovery was 99.2%(RSD=0.84%, n=9). With confidence probability (P)95%, the expanded uncertainty of 2, 3, 5, 4′-tetrahydroxystilbene-2-O-β-D-glucos ide in processed polygonum multiflorum was 0.02%. Conclusion: The uncertainty analysis method is applicable to evaluation of uncertainty in measurement of 2, 3, 5, 4′-tetrahydroxystilbene-2-O-β-D-glucos ide in processed polygonum multiflorum.
