四黄栓药材浸膏中盐酸小檗碱的含量测定和栓剂成型工艺研究

摘要: 目的:探索院内制剂四黄栓药材浸膏中盐酸小檗碱含量的测定方法,并对栓剂成型工艺进行优化。方法: 采用HPLC定量测定法,探索药材浸膏中盐酸小檗碱含量的测定方法;设计正交试验,以基质熔融温度、含药基质灌模温度以及脱模时间为考察因素,以栓剂外观性状、装量差异及融变时限为综合指标,优选出四黄栓最佳成型工艺。结果: 四黄栓药材浸膏中盐酸小檗碱含量的测定方法为:采用Phenomenex C18柱(4.6 mm × 250 mm,5 μm);以乙腈-0.05 mol·L-1磷酸二氢钠溶液(50∶50) 为流动相(每100 mL中加十二烷基硫酸钠0.4 g,再以磷酸调节pH值为4.0);检测波长为345 nm(VWD紫外检测器);进样量为10 μL;流速为0.8 mL·min-1;柱温为25 ℃。最佳成型工艺为将基质在70 ℃时水浴加热完全熔融后,加入研细过100目筛的浸膏粉,搅拌均匀,冷却至温度约40 ℃时,灌注于栓模内,固化,20 min后脱模,即得。结论: 初步建立了四黄栓药材浸膏中盐酸小檗碱的含量测定方法,该检测方法稳定性好、重复性好;优选出了四黄栓成型工艺,该工艺简单合理、稳定可行。

Abstract: Objective: To explore the method for determining the content of berberine hydrochloride in the extract of sihuang suppository, and to optimize the molding process of the suppository. Methods: HPLC quantitative determination was used to explore the method for determining the content of berberine hydrochloride in the extract. Orthogonal experiments were designed, and the melting temperature of the matrix, the filling temperature of the drug-containing matrix and the demolding time were used as the investigating factors. The appearance, properties, weight variation, melting time limit were used as comprehensive indicators, and the best molding process of sihuang suppository was selected. Results: The method for determining the content of berberine hydrochloride in sihuang suppository extract is as follows: Phenomenex C18 column (4.6 mm×250 mm, 5 μm), acetonitrile- 0.05 mol·L-1 sodium dihydrogen phosphate solution (50∶50) as the mobile phase (add 0.4 g sodium dodecyl sulfate per 100 mL, and adjust the pH to 4.0 with phosphoric acid). The detection wavelength is 345 nm (VWD ultraviolet detector). The injection volume is 10 μL.The flow rate is 0.8 mL·min-1. The column temperature is 25 ℃. The best molding process is to heat the matrix in a water bath at 70 ℃ to completely melt it, add the finely ground extract powder that has passed through a 100-mesh sieve, and stir evenly. Cool to a temperature of about 40 ℃, pour it into the suppository mold, cure for 20 minutes, and release the mold to get it. Conclusion: A method for the determination of berberine hydrochloride content in sihuang suppository extract was preliminarily established, and the detection method has good stability and reproducibility. The molding process of sihuang suppository was optimized, which is simple, reasonable, stable and feasible.
