清胰汤治疗胰腺炎的网络药理学研究

Network Pharmacology Research on Qingyi Deoction Treating Pancreatitis

摘要: 目的:基于网络药理学的方法探讨清胰汤治疗胰腺炎(pancreatitis)的作用机制。方法:通过中药系统药理学数据库(TCMSP)及中药整合药理学研究平台(TCMIP)检索相关文献,获取清胰汤所含中药的主要化学成分,收集其相对应的靶点;结合Cytoscape 3.8.0构建有效成分-核心靶点网络,靶标蛋白相互作用(PPI)网络。利用Bioconductor中的R包对核心靶标进行GO富集和KEGG通路分析。结果:共获得清胰汤药物化学成分105个,靶点250个;获得疾病靶点289个,其中核心靶标25个;核心蛋白涉及EGFR、AKT1、IL6等,通路主要为PI3K-Akt、AGE-RAGE、HIF-1等炎症通路和肿瘤蛋白多糖、结直肠癌、膀胱癌等癌症通路;处方中关键的药物是延胡索、大黄、白芍、柴胡与黄芩,与靶点关系最紧密的成分为槲皮素、β-谷甾醇、山奈酚、汉黄芩素、金合欢素、黄芩素和芦荟大黄素。结论:本研究初步展示了清胰汤治疗胰腺炎的多成分、多靶点、多通路的作用机制,为后期药物分离纯化及药理研究提供了基础,筛选出的作用靶点和通路对于诊断和治疗胰腺炎有一定的参考价值。同时提示对癌症的治疗可以借鉴清胰汤的治疗思路。

Abstract: Objective:To investigate the mechanism of qingyi deoction treating pancreatitis based on the method of network pharmacology. Methods: Relevant literatures were retrieved through the Chinese Medicine System Pharmacology Database (TCMSP),Chinese Medicine Integrated Pharmacology Research Platform (TCMIP). The main chemical compositions of the Chinese herbal medicine contained in qingyi deoction were obtained, and the corresponding targets were collected. Combined with Cytoscape 3.8.0, the active ingredient-core target network and target protein interaction PPI network were constructed. R software package in Bioconductor was used for the GO enrichment and KEGG pathway analysis. Results: There were 105 chemical components in qingyi deoction, and 250 targets were found. 289 disease targets were obtained, including 25 core targets. The core proteins involved EGFR, AKT1, IL-6, etc., and the pathway mainly included PI3k-Akt, AGE-RAGE, HIF-1 inflammation pathway and tumor proteoglycan, colorectal cancer, bladder cancer and other cancer pathways. The key Chinese herbal medicines in qingyi deoction were rhizoma corydalis, rhubarb, white paeony root, radix bupleuri,and scutellaria baicalensis. The important target components included quercetin, beta-sitosterol, kaempferol, wogonin,acacetin, baicalein and aloe-emodin. Conclusion: The study preliminarily demonstrated the multi-component, multi-target and multi-channel action mechanism of qingyi deoction in the treatment of pancreatitis,and provided a foundation for the later separation and purification and pharmacological study. The target and pathway screened out have certain reference value for the diagnosis and treatment of pancreatitis. At the same time, it was suggested that the treatment ideas of qingyi deoction can be used for reference in the treatment of cancer.
