不同评价方案对校准品互换性评估的差异研究及对注册技术审评工作的启示

Research on the Differences of the Commutability Evaluation of Calibrators by Different Evaluation Schemes and Its Implication for Technical Review of the Registration

摘要: 目的:分析2个不同评价方案对校准品互换性评估的差异。方法:以B型利钠肽工作校准品为例,分别使用《WS/T 3562011基质效应与互通性评估指南》和《EP 14A3 评价处理后样品的互换性:第三版核准指南》2个不同文件推荐的评价方案,对10个B型利钠肽工作校准品进行互换性评估,并比较不同方案互换性评估的差异。结果:两个评价方案对B型利钠肽校准品互换性评估结果不同,但最终结论一致,即所有校准品的测量值均落在95%预测区间内,所评估校准品均具有互换性。结论:评价方案不同,相同制备样品的互换性评估结论可能相同,也可能不同。EP 14A3指南的互换性评价方案更符合医学预期需求。同时,技术审评机构需对校准品互换性研究给予更多关注。

Abstract: Objective: To analyse the difference of the two different evaluation schemes on the commutability evaluation of calibrators. Methods:Btype natriuretic peptide(BNP) working calibrator were taken as an example, and the commutability of the treated samples were evaluated by using the two evalution schemes from WS/T3562011 and EP14A3 respectively. The commutability of 10 BNP working calibrators was evaluated and the differences of the two schemes were compared. Results:The results of commutability evaluation of BNP calibrators were different between the two evaluation schemes, but the final conclusions were the same (all the calibrators evaluated were commutable since the measured values were all in 95% prediction interval). Conclusion: The commutability evaluation conclusions of the same processed sample may be same or different for different evaluation schemes. The commutability evaluation scheme of EP14A3 guidelines is more in line with medical expectations. Moreover, technical review institutions should pay more attention to the commutability evaluation of calibrators.
