橘红颗粒微生物限度检查方法适用性研究

Applicability of Microbial Limit Test Method of Juhong Granules

摘要: 目的:建立橘红颗粒的微生物限度检查方法。方法:依据2020年版《中华人民共和国药典》四部通则1105、1106、1107,进行了微生物限度检查方法的适用性研究。结果:采用平皿法和薄膜过滤法进行了需氧菌总数方法适用性试验,采用平皿法开展了霉菌和酵母菌总数方法适用性试验,回收比值均在0.5~2.0,表明该药品对阳性试验菌均无抑菌性;采用常规法和增加培养基体积法进行了控制菌(大肠埃希菌)检查方法适用性试验,试验组和阳性对照组均检出了阳性菌,表明该药品对大肠埃希菌无抑菌性。结论:可采用平皿法(1∶10)进行需氧菌总数及霉菌和酵母菌总数的检查,采用常规法进行控制菌的检查。该方法易操作、重复性好,可用于橘红颗粒的微生物限度检查。

Abstract: Objective:To establish the microbial limit test method of juhong granules. Methods: According to the general rules 1105, 1106 and 1107 of the fourth part of Chinese Pharmacopoeia (2020 edition), the applicability of the microbial limit test method was studied. Results: The plate method and membrane filtration method were used to carry out the applicability test of aerobic bacteria count method, and the plate method was used to carry out the applicability test of mold and yeast count method. The microbial recovery ratio was 0.5-2.0, indicating the drug had no bacteriostasis to the positive test bacteria. The applicability test of the control bacteria (Escherichia coli) inspection method was conducted by using the conventional method and the method of increasing the volume of culture medium. Positive bacteria were detected in both the test group and the positive control group, indicating the drug has no bacteriostasis to Escherichia coli. Conclusion: The plate method (1∶10) can be used to check the total number of aerobic bacteria, mold and yeast, and the routine method can be used to check the control bacteria. The method is easy to operate and has good repeatability, which can be used for microbial limit test of juhong granules.
