古代经典名方升陷汤治疗心血管疾病的作用机制和物质基准指标成分研究

Study on the Action Mechanism and Material Reference Index Composition of Ancient Classical Prescription Shengxian Decoction in the Treatment of Cardiovascular Disease

摘要: 目的:探讨古代经典名方升陷汤用于心血管疾病治疗的作用机制与药效物质基础。方法: 运用网络药理学方法,采用多个数据库收集的升陷汤中化学成分信息,预测化学成分的作用靶点,通过Cytoscape软件,构建活性成分-靶点相互作用和靶点蛋白相互作用网络,通过DAVID数据库对靶点进行生物过程和信号通路分析,针对心血管疾病构建“药材-活性成分-靶点-通路”网络。结果: 升陷汤5种药材中的44个潜在活性成分,通过作用于72个靶点,影响9个心血管相关的基因通路,发挥治疗心血管相关疾病的作用。经分子对接验证,可以从中确定为物质基准质控指标的核心成分共10个,发挥作用的关键核心靶点9个。结论:升陷汤用于心血管疾病治疗具有多成分、多靶点、多通路的特点,采用网络药理学方法可以实现快速筛选活性成分预测作用机制目的,可为古代经典名方物质基准指标成分的确定提供依据。

Abstract: Objective:To explore the action mechanism and pharmacodynamic substance basis of ancient classical prescription sheng-xian decoction in the treatment of cardiovascular disease. Methods:Using the method of network pharmacology, the information of chemical components in shengxia decoction were collected from multiple databases, and the targets of chemical components were predicted. The compounds-targets network was constructed by Cytoscape software and network topology analysis was carried out. DAVID database was used to analyze the molecular function and biological pathway of the action targets. "herbs-active compounds-targets-pathways" network was constructed for cardiovascular diseases. Results:44 potential active compounds were screened out from 5 kinds of herbs in shengxian decoction, which can affect 9 cardiovascular related gene pathways by acting on 72 targets, and play a role in the treatment of cardiovascular related diseases. Through molecular docking verification, there are 10 core components that can be determined as the material benchmark quality control indicators, and 9 key targets. Conclusion:Shengxian decoction can treat cardiovascular disease through multi-target, multi-channel and multi-pathway synergism. The network pharmacology method can be used to quickly screen the active ingredients and predict the mechanism of action, and provide the basis for determining the material benchmark indicators of ancient classical prescription.
