HPLC-DAD法同时测定延胡索配方颗粒中10个标志性成分

Simultaneous Determination of Ten Marked Components in Yanhusuo Formula Granules by HPLC-DAD

摘要: 目的:建立同时测定延胡索配方颗粒中四氢非洲防己碱、D-四氢药根碱、原阿片碱、延胡索乙素、黄连碱、四氢小檗碱、延胡索甲素、盐酸巴马汀、盐酸小檗碱、去氢延胡索甲素10个生物碱成分含量的高效液相色谱测定方法。方法: 采用Agilent ZORBAX SB-C18色谱柱(4.6 mm×250 mm,5 μm),以乙腈-0.1%磷酸溶液(含0.08%三乙胺)为流动相,梯度洗脱,检测波长280 nm,流速1.0 mL·min-1,柱温30℃。结果:10个成分在上述条件下均达到基线分离,线性关系良好(r >0.999 5,n=5);平均加样回收率为96.90%~100.05%,RSD为0.7%~1.9%(n=6),8批延胡索配方颗粒样品中上述10个标志性成分的平均含量分别为0.740、0.488、0.973、7.227、1.242、1.599、1.513、0.720、0.218、4.479 mg·g-1。结论:本方法操作简便、准确、重复性好,可用于延胡索配方颗粒多成分含量的同时测定,为延胡索配方颗粒质量标准提升提供参考依据。

Abstract: Objective:To establish the HPLC method for simultaneous determination of the content of tetrahydrocolumbamine, corypalmine, protopine, tetrahydropalmatine, coptisine, tetrahydroberberine, corydaline, palmatine hydrochloride, berberine hydrochloride, dehydrocorydaline in yanhusuo fomula granules. Methods:The analysis was performed on Agilent ZORBAX SB-C18 column (4.6 mm×250 mm, 5 μm) by gradient elution of acetonitrile-0.1% phosphoric acid (containing 0.08% triethylamine). The UV detection wavelength was set at 280 nm, and the flow rate was 1.0 mL·min-1. The column temperature was 30℃. Results:The 10 components reached baseline separation under the above conditions, and the linear relationship was good (r>0.999 5, n=5). The average recoveries (n=6) were 96.90%-100.05% (RSD=0.7%-1.9%).The average contents of the 10 components in 8 batches of yanhusuo formula granules were 0.740, 0.488, 0.973, 7.227, 1.242, 1.599, 1.513, 0.720, 0.218, 4.479 mg·g-1, respectively. Conclusion: The method is simple, accurate and reproducible, and can be used for the determination of multiple components in yanhusuo fomula granules, which can provide reference for the comprehensive evaluation of the quality.
