基于miRNA的中药药效量化策略

摘要/Abstract

摘要： 中药是动植物共进化的产物。中药中的天然产物绝大部分是人体基因表达的调节剂，这些天然产物主要通过综合协同效应和组合效应等多种模式影响和调节人体各器官的基因表达谱，进而起到治疗疾病的作用。因此，人体基因活动的变化可以量化中药药效。但由于人体的基因调控呈网络结构，且中药中所含天然产物高度复杂，对人体基因活动的表征势必需要多指标体系。本研究从进化生物学视角，基于可操作性，综合分析认为miRNA是目前最适合用来表征人体基因活动状况的生物标志物，通过“三步走”策略，构建中药药效量化的“Q-marker”体系，以期为实现中药药效量化，用天然产物作为“Q-marker”全面质控中药提供新的思路，为中药质量标志物“Q-marker”的推进起到建设性作用。

Abstract: Traditional Chinese medicine（TCM）is a product of co-evolution of animals and plants. Most of the natural products in TCM are the regulators of gene expression in the human body. These natural products affect and regulate the gene expression profile of the human body through multiple approaches such as comprehensive synergistic effects and combination effects， and then play the role of disease treatment. Therefore， it is possible to quantify the drug efficacy of TCM through characterizing the changes in gene expression caused by TCM. However， due to the network-structure of human gene expression regulation and the complexity of natural products contained in TCM， it is essential to use a multi-biomarker system for the characterization of gene expression status. This study attempts to explore from the perspective of evolutionary biology，and based on the operability，and it is believed that miRNAs are currently the most suitable biomarkers to characterize human gene activity.It is possible to construct a "Q-marker" system for quantifying the efficacy of TCM， through the "three-step" strategy，in order to quantify the efficacy of TCM，and to use natural products as "Q-marker" for comprehensive quality control of TCM， which plays a constructive role in the promotion of TCM quality marker "Q-marker".
