华蟾素注射液联合5-氟尿嘧啶对大肠癌LoVo细胞粘附侵袭能力的影响

摘要: 目的:探讨华蟾素注射液联合5-氟尿嘧啶(5-Fu)对于大肠癌LoVo细胞粘附侵袭能力的影响。方法:根据实验目的将大肠癌LoVo细胞划分为如下4组:空白对照组、5-Fu组、华蟾素组、5-Fu+华蟾素组。通过CCK-8法检测细胞粘附能力,通过Transwell小室法对细胞侵袭能力加以检验。结果:通过测定各组在450 nm波长处的吸光值(OD450值)得到:5-Fu+华蟾素组的OD450值低于其余3组,5-Fu组的OD450值低于对照组和华蟾素组,华蟾素组的OD450值低于对照组(P<0.01);通过测定各组细胞侵袭个数得出:5-Fu+华蟾素组LoVo细胞侵袭细胞数目低于其余三组,5-Fu组LoVo细胞侵袭细胞数目低于对照组和华蟾素组,华蟾素组LoVo细胞侵袭细胞数目低于对照组(P<0.01)。结论:5-Fu+华蟾素可以有效抑制大肠癌LoVo细胞的粘附侵袭能力,联合用药较单药使用效果更显著,但是常规化疗治疗手段优于单独使用华蟾素。

Abstract: Objective:To investigate the effect of cinobufacini injection combined with 5-fluorouracil (5-Fu) on the adhesion and invasion ability of colorectal carcinoma LoVo cells. Methods:According to the purpose of the experiment, colorectal carcinoma LoVo cells were divided into the following four groups:the control group, the 5-Fu group, the cinobufacini group, and the 5-Fu + cinobufacini group. The cell adhesion ability was detected by CCK-8 method and the cell invasion ability was detected by Transwell chamber method. Results:By measuring absorbance at 450 nm (OD450) in each group, it was found that the OD450 of 5-Fu+ cinobufacini group was lower than that of the other three groups, the OD450 of 5-Fu group was lower than that of the control group and cinobufacini group, and the OD450 of cinobufacini group was lower than that of the control group(P< 0.01). By measuring the number of invasive cells in each group, it was found that the number of invasive cells of LoVo cells in 5-Fu+ cinobufacini group was lower than that in the other three groups, the number of invasive cells of LoVo cells in 5-Fu group was lower than that in the control group and the cinobufacini group, and the number of invasive cells of LoVo cells in cinobufacini group was lower than that in the control group (P < 0.01). Conclusion:5-Fu + cinobufacini can effectively inhibit the adhesion and invasionability of colorectal carcinoma LoVo cells, and the inhibit effect of combined medication of 5-Fu and cinobufacini is more significant than sigle drug use.
