稳心安神颗粒HPLC指纹图谱的建立及6种成分测定

Establishment of HPLC Fingerprint and Determination of Six Components of Wenxin Anshen Granules

摘要: 目的:建立稳心安神颗粒的高效液相色谱(HPLC)指纹图谱,并同时测定2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷、盐酸小檗碱、丹酚酸B、槲皮素、五味子醇甲、丹参酮Ⅱ A的含量。方法:采用高效液相色谱法,Alphasil VC-C18(4.6 mm×250 mm,5 μm)色谱柱,流动相:乙腈-0.1%磷酸溶液,梯度洗脱,流速为0.9 mL·min-1,检测波长为280 nm,进样量为10 μL,柱温为25 ℃;建立指纹图谱,采用国家药典委员会“中药色谱指纹图谱相似度评价系统(2012.130723版)”对10批稳心安神颗粒进行相似度评价,对2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷、盐酸小檗碱、丹酚酸B、槲皮素、五味子醇甲、丹参酮ⅡA色谱峰进行指认,同时测定6种成分的含量。结果:10批稳心安神颗粒中标定25个共有峰,相似度均在0.950以上。6个成分分别在0.021 0~0.210 0、0.416 8~4.168 0、0.404 4~4.044 0、0.010 2~0.102 0、0.042 6~0.426 0、0.044 8~0.448 0 μg进样量范围内线性关系良好,平均回收率为98.61%~100.69%,RSD为0.75%~1.26%。结论:该方法重复性好,可操作性强,可用于控制稳心安神颗粒的质量。

Abstract: Objective:To establish the HPLC fingerprint of wenxin anshen granules, and simultaneously determine the contents of 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside,berberine hydrochloride, salvianolic acid B, quercetin, schisandrin A and tanshinone IIA. Methods:HPLC was used. Alphasil VC-C18(4.6 mm×250 mm,5 μm)chromatographic column, with the mobile phase of acetonitrile-0.1% phosphoric acid,gradient elution at a flow rate of 0.9 mL·min-1. The detection wavelength was 280 nm, the injection volume was 10 μL, and the column temperature was 25 ℃.The fingerprint spectrum was established and the similarity of 10 batches of wenxin anshen granules was evaluated by using the similarity evaluation system of Chromatographic Fingerprint of Traditional Chinese Medicine (version 2012.130723). The Chromatographic peaks of 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside, berberine hydrochloride, salvianolic acid B, quercetin, schisandrin A and tanshinone IIA were identified and the content of the 6 components were determined. Results:25 common peaks were identified in 10 batches of wenxin anshen granules, and the similarities were all above 0.950. The linear ranges of the six components were 0.021 0-0.210 0、0.416 8-4.168 0、0.404 4-4.044 0、0.010 2-0.102 0、0.042 6-0.426 0、0.044 8-0.448 0 μg respectively, and the average recoveries was 98.61%-100.69%,RSD was 0.75%-1.26%. Conclusion:The method has good repeatability and strong operability, and can be used to control the quality of wenxin anshen granules.
