HPLC法同时测定复方硫酸软骨素片中3个活性成分的含量

Simultaneous Determination of Three Active Components in Compound Chondroitin Sulfate Tablets by HPLC

摘要: 目的:建立HPLC梯度洗脱法同时测定复方硫酸软骨素片中硫酸软骨素、芍药苷和甘草酸3个活性成分的含量。方法:采用InertSustain C18色谱柱(4.6 mm×250 mm,5 μm),以5 mmol·L-1庚烷磺酸钠溶液为流动相A,乙腈为流动相B,梯度洗脱,柱温为30 ℃,检测波长为230 nm。结果:硫酸软骨素、芍药苷和甘草酸分别在0.292 3~7.307 6、0.002 6~0.132 0和0.006 1~0.301 8 mg·mL-1范围内与峰面积呈良好的线性关系;平均加样回收率为98.5%~99.7%,RSD为1.0%~1.2%。4批样品中,硫酸软骨素、芍药苷和甘草酸的含量分别为72.954 7~74.103 6、0.661 3~0.689 8和1.502 3~1.520 9 mg/片。结论:本研究建立HPLC梯度洗脱法同时测定复方硫酸软骨素片中3个活性成分,该方法简便、快速、准确,可用于复方硫酸软骨素片的质量控制。

Abstract: Objective: To establish an HPLC gradient elution method for simultaneous determination of chondroitin sulfate, paeoniflorin and glycyrrhizic acid in compound chondroitin sulfate tablets. Methods: The chromatographic separation was performed on InertSustain C18 column(4.6 mm×250 mm,5 μm),and gradient elution was conducted with 5 mmol·L-1 sodium heptanesulfonate solution as mobile phase A and acetonitrile as mobile phase B. The column temperature was 30℃, and the detection wavelength was 230nm. Results: The linear ranges of chondroitin sulfate, paeoniflorin and glycyrrhizic acid were 0.292 3-7.307 6, 0.002 6-0.132 0 and 0.006 1-0.301 8 mg·mL-1, respectively. The average recoveries were 98.5%-99.7%, and RSD was 1.0%-1.2%. The content ranges of chondroitin sulfate, paeoniflorin and glycyrrhizic acid in 4 batches of samples were 72.954 7-74.103 6, 0.661 3-0.689 8 and 1.502 3-1.520 9 mg per tablet, respectively. Conclusion: The HPLC gradient elution method has been established for simultaneous determination of the contents of three active components in compound chondroitin sulfate tablets. The method is simple, rapid and accurate, and suitable to be used for the quality control of compound chondroitin sulfate tablets.
