贵州望谟县种植基地4种生态型艾纳香药材中3种成分的含量测定

Content Determination of 3 Components in 4 Ecotypes of Blumea balsamifera Planted in Guizhou Wangmo County

摘要: 目的:采用气相色谱(GC)法建立艾纳香药材中樟脑、左旋龙脑(L-龙脑)、β-石竹烯的含量测定方法,并用于测定贵州望谟县种植基地4种生态型5个采收期的艾纳香药材含量。方法:以HP-5毛细管柱(30 m×0.32 mm,0.25 μm)进行测定,程序升温,进样口温度为200 ℃,检测器温度为280 ℃,进样量为1 μL,分流进样。结果和结论:该方法简单、准确、重复性好,可用于测定艾纳香中樟脑、左旋龙脑(L-龙脑)、β-石竹烯的含量。3种成分在不同生态型的不同采收期艾纳香药材中的含量有差异,其中L-龙脑在马耳艾和大叶艾中含量高于其他2种生态型;樟脑在大叶艾含量较低,在马耳艾含量较高;β-石竹烯在野艾和小叶艾中的含量高于其他2种生态型。根据《中华人民共和国药典》艾片的质量要求,栽培艾纳香作为提取艾粉原料药材时,优先考虑栽培大叶艾,且在11月下旬时便可开始收集进行提取。

Abstract: Objective: To establish a GC method for content determination of bornanone, L-borneol and β-caryophyllene in Blumea balsamifera (B. balsamifera), and to determine the contents of the above three components in 4 ecotypes of B. balsamifera at five life stage which was planted in Guizhou Wangmo county. Methods: The chromatographic separation was performed on HP-5 capillary column(30 m×0.32 mm, 0.25 μm), temperature programmed, and the inlet temperature was set at 200 ℃. The temperature of the detector was controlled at 280 ℃ and the injection volume was 1 μL with split injection. Results and Conclusion: The established method is sample, accurate and repeatable, and can be used for content determination of bornanone, L-borneol and β-caryophyllene in B. balsamifera. The contents of three component were varied in different ecotypes at various life stages. Contents of L-borneol “in horse” ear type and large-leaf type were higher than other two ecotypes. Contents of bornanone was lower in lobular type, while higher “in horse” ear type. Contents of β-caryophyllene was higher in wild type and lobular type than in other two ecotypes. According to the quality requirements of Pharmacopoeia of the People′s Republic of China, it is suggested that when use B. balsamifera as the raw material to extract sage parder, it is preferable to cultivate large-leaf type, and it should be collected for extraction in late November.
