血虚证动物模型建立及评价指标研究

摘要:

血虚证是中医临床上常见的证型之一,由于人体中的血液无法滋养五脏六腑、经脉、四肢百骸,临床表现为面色无华、头晕目眩、心慌失眠等。临床上对于血虚证的诊断很大程度受医者个人经验和主观判断影响,使得血虚证的诊断难以统一和标准化,导致血虚证研究发展滞后。为推动血虚证的临床诊断标准化及研究进程,建立科学、合理、与中医临床实践相符合的血虚证动物模型及其评价体系尤为重要。血虚证动物模型的建立和评价指标的研究为中医药治疗血虚证提供了重要的实验基础。目前,血虚证动物模型主要通过物理、化学等手段损耗实验动物血液或抑制其造血功能来制备。本研究总结血虚证动物模型建立的常用方法,包括失血法、化学损伤法、溶血法、辐射损伤法和复合药物法,根据血虚证临床患者的症状,分析血虚证动物模型的评价指标,如一般体征、外周血象、骨髓造血功能、造血及免疫因子和多组学的生物标志物,以期提高后续血虚证相关研究的科学性和临床应用价值。

Abstract:

 Blood deficiency syndrome is one of the most common clinical patterns in Traditional Chinese Medicine TCM. It arises when insufficient blood fails to nourish the internal organs meridians and extremities manifesting clinically as pallor dizziness palpitations insomnia and other symptoms. However the diagnosis of blood deficiency syndrome in clinical practice remains heavily reliant on physicians' subjective judgment and individual experience leading to inconsistencies in diagnostic criteria and hindering standardized research progress. To advance the standardization of clinical diagnosis and research on blood deficiency syndrome it is crucial to establish scientifically robust and clinically relevant animal models that align with TCM principlesalong with a systematic evaluation framework. The development of these models and their evaluation metrics has laid an essential experimental foundation for TCM-based therapies targeting blood deficiency syndrome. Animal models of blood deficiency syndrome are primarily established through physical or chemical interventions that deplete blood or suppress hematopoietic function. This study summarizes common methodologies for constructing blood deficiency syndrome animal models including blood loss chemical injury hemolysis radiation injury and compound drug administration. Furthermore based on the clinical symptoms observed in patients this review analyzes evaluation indicators for these models such as general physical signs peripheral blood parameters bone marrow hematopoietic function hematopoietic/immune factors and multi-omics biomarkers. These insights aim to enhance the scientific rigor and clinical applicability of future research on blood deficiency syndrome.
