APTT法测定注射用重组人凝血因子Ⅷ中人凝血因子Ⅷ含量的不确定度评定

Uncertainty Assessment of Determination of Human Coagulation Factor Content in Recombinant Human Coagulation Factor for Injection by APTT Method

摘要:

目的:建立测定注射用重组人凝血因子中人凝血因子含量不确定度的评定方法。方法:采用进口注册标准JS20210052中人凝血因子含量测定方法进行测定,同时依据《测量不确定度评定与表示》JJF1059.1-2012中的相关规定,建立测定人凝血因子的不确定度数学模型,并对不确定度来源进行分析和评定。结果:人凝血因子含量的不确定度来源主要有对照品溶液制备(CV)、供试品溶液制备(VS)、供试品测量重复性(SS)和对照品标准曲线(Ci)、活化部分凝血活酶时间(APTT)法(以下简称APTT法)测定注射用重组人凝血因子中人凝血因子含量的合成不确定度为0.880 17 IU·mL-1,扩展不确定度为1.760 34k=2,置信概率为95%)。结论:注射用重组人凝血因子的不确定度贡献主要来源于供试品溶液的配制及标准品的标准曲线制备,而对照品称量和配制的不确定度在本实验中贡献不大,可忽略不计。本研究量化了此测定分析方法的不确定度,所建立的方法适用于APTT法测定注射用重组人凝血因子中人凝血因子含量的不确定度评定。

Abstract:

Objective: To establish an evaluation method for the uncertainty of human coagulation factor Ⅷ content in recombinant human coagulation factor Ⅷ for injection.Method: The determination of human coagulation factor Ⅷ content was carried out using the import registration standard JS20210052. At the same time according to the relevant provisions of JJF1059.1-2012 "Evaluation and Expression of Uncertainty in Measurement" a mathematical model of uncertainty for the determination of human coagulation factor Ⅷ was established and the sources of uncertainty were analyzed and evaluated.Results: The sources of uncertainty in the content of human coagulation factor Ⅷ mainly include preparation of reference solution CV), preparation of test solution VS), measurement repeatability of test sample SS and standard curve of reference sample Ci. The combined uncertainty of the determination of human coagulation factor Ⅷ content in recombinant human coagulation factor Ⅷ for injection by activated partial thromboplastin time APTT method is 0.880 17 IU·mL-1 and the expanded uncertainty is 1.760 34 k=2 confidence probability is 95%.Conclusion: The contribution to the uncertainty in determination of human coagulation factor Ⅷ contene in recombinant human coagulation factor Ⅷ for injection mainly comes from the preparation of the test solution and the standard curve of the standard substance while the uncertainty contribution of the weighing and preparation of the reference sample is not significant in this experiment and can be ignored. This study quantified the uncertainty of this analytical method and the established method is suitable for evaluating the uncertainty of the APTT method for determining the content of human coagulation factor Ⅷ in recombinant human coagulation factor Ⅷ for injection.
