三七治疗骨质疏松性骨折的网络药理学分析

Network Pharmacologic Analysis of Panax notoginseng in the Treatment of Osteoporotic Fractures

摘要:

目的:基于网络药理学的理论框架,深入探讨三七治疗骨质疏松性骨折发挥作用的机制。方法:利用TCMSP数据库对三七主要活性成分和作用靶点进行筛选,应用GeneCardsOMIM数据库对骨质疏松性骨折相关靶点进行预测,使用微生信在线平台制作维恩图,利用Cytoscape软件绘制中药-成分-疾病-靶点调控网络获取共同靶点。基因本体富集分析(GO)以及基因相互作用通路分析(KEGG)是通过R语言和 Bioconductor 平台来进行。结果:三七对骨质疏松性骨折的治疗主要是通过7种有效成分,128个作用靶点起作用。其中主要活性成分为β-谷甾醇、豆甾醇、槲皮素、人参皂苷Rh2等;主要核心靶点有AKT1IL6TNFTP53等;重点通道有动脉粥样硬化信号通道,大肠癌信号通道,肿瘤蛋白多糖信号通道,白介素-17信号通道,TNF信号通道等。结论:网络药理学揭示了三七在治疗性骨质疏松骨折方面的特性,这些特性体现在其多成分、多靶点和多通路的作用机制上,为后续研究奠定了基础。

Abstract:

Objective: Based on network pharmacology to explore the mechanisms of Panax notoginseng in the treatment osteoporotic fractures.Methods: Using the TCMSP database to identify active ingredients and targets of Panax notoginseng. GeneCards and OMIM predicted osteoporotic fracture-related targets. Venn diagrams were generated via an online bioinformatics platformand Cytoscape mapped TCM-ingredient-disease target networks. Results: The treatment of osteoporotic fractures by Panax notoginseng mainly relies on seven active ingredients β-sitosterolstigmasterolquercetinginsenoside Rh2 etc. and 128 targets AKT1 IL6 TNF TP53 etc.. Key pathways include atherosclerosis signaling pathway tumor proteoglycan signaling pathway colorectal cancer signaling pathwayinterleukin-17 signaling pathway and TNF signaling.Conclusion: Network pharmacology revealed that Panax notoginseng treats osteoporotic fractures through multi-componentmulti-targetand multi-pathway mechanismssupporting future research.
