miR-381 在HPV感染型宫颈癌中的表达及对宫颈癌细胞增殖、迁移和侵袭的影响

摘要: 目的: 探讨miR-381在人乳头瘤病毒(HPV)感染型宫颈癌中的表达,以及对HPV感染型宫颈癌细胞增殖、迁移和侵袭的影响。方法:收集HPV感染型宫颈癌组织12例,通过q-PCR法检测miR-381的表达。将宫颈癌细胞C33A、HCE1随机分为5组,一组Control,一组miR-381 mimics NC,一组miR-381 mimics,一组miR-381 inhibitor NC,一组miR-381 inhibitor。采用CCK-8细胞增殖实验检测细胞增殖能力,采用细胞划痕实验检测细胞迁移能力,采用细胞侵袭实验检测细胞侵袭能力。结果:miR-381在HPV感染型宫颈癌组织和细胞系中表达均降低(P<0.05)。在宫颈癌C33A和HCE1细胞中,miR-381 mimics组细胞增殖、迁移和侵袭能力均降低(P<0.05),miR-381 inhibitor组细胞增殖、迁移和侵袭能力均增加(P<0.05)。结论: miR-381 在HPV感染型宫颈癌中呈低表达,可能通过靶向调节抑制HPV感染型宫颈癌细胞的增殖、迁移及侵袭能力。

Abstract: Objective: To study the expression of miR-381 in HPV infection cervical cancer, and its effect on HPV infection cervical cancer cell proliferation,migration and invasion. Methods: Collect 12 cases of HPV infection cervical cancer tissue, and use q-PCR method to detect the expression of miR-381. Each type of HPV infection cervical cells were randomly divided into five groups: a group was not transfected as control, a group of transfection with miR-381 mimics NC, a group of transfection with miR-381 mimics, a group of transfection with miR-381 inhibitor NC, and a group of transfection with miR-381 inhibitor.The CCK 8 experimental was use to detect cell proliferation ability, the cell scratch test was use to detect cell migration ability,and the cell invasion test was use to detect cell invasion ability. Results: miR-381 expression in HPV infection cervical cancer tissues and cell lines were lower( P<0. 05). In C33A and HCE1 cells, miR-381 mimics cell proliferation, migration and invasion ability were lower (P<0.05). miR-381 inhibitor cell proliferation, migration and invasion ability were increased(P<0.05). Conclusion:The low expression of miR-381 in HPV infection cervical cancer, possibly through targeted adjustment inhibition of HPV infection cervical cancer cell proliferation, migration and invasion ability.
