细胞凋亡核心基因Caspase家族的研究进展

摘要: 细胞凋亡(apoptosis),又称程序性细胞死亡(PCD),是为了维持机体内环境的稳定,细胞内预存的由相关基因控制的死亡程序被某种因素诱发,进而引起细胞死亡的过程。细胞凋亡在人体的胚胎发生、神经系统的功能完善、免疫功能的调节、疾病的发生发展过程中,均发挥着至关重要的作用。Caspase是一种参与细胞凋亡的半胱氨酸蛋白酶,它的家族成员直接或间接的引起细胞凋亡结构特征的出现,处于细胞凋亡传导通路的核心位置。Caspase家族成员通过启动外源性细胞凋亡传导通路及间接启动内源性细胞凋亡传导通路,在细胞凋亡的整个过程中发挥关键作用。但目前为止,Caspase家族成员的具体作用机制、调控机制、与疾病的具体关系尚未完全明确,因此对其深入研究将对细胞凋亡相关疾病及肿瘤发病机制的阐明提供重要依据,从而可能为临床疾病治疗提供新途径。本文拟对Caspase分类、Caspase凋亡传导通路、Caspase的调控及Caspase与肿瘤的发生等相关研究进展做一简要综述,以期提供借鉴。

Abstract: Apoptosis, also known as programmed cell death (PCD), is a process in which the pre-existing death program controlled by related genes in the cell induced by certain factors, which in turn causes cell death,in order to maintain the stability of the environment in the body.Apoptosis plays a vital role in the human embryogenesis, the perfection of the nervous system, the regulation of immune function, and the occurrence and development of diseases. Caspase is a cysteine protease involved in apoptosis. Its family members directly or indirectly cause the appearance of structural features of apoptosis. Caspase are at the core of the apoptosis conduction pathway.Caspase family members play a key role in the entire process of cell apoptosis by initiating the exogenous apoptosis pathway and indirectly initiating the endogenous apoptosis pathway.However, so far, the specific mechanism of action, regulation mechanism, and specific relationship between Caspase family members and the disease have not been completely cleared. Therefore, in-depth study will provide an important basis for the elucidation of apoptosis-related diseases and tumor pathogenesis, and then may provide a new way for the treatment of clinical diseases. In this article, brief review are made about the research progress of Caspase family, Caspase apoptosis pathway, Caspase regulation and Caspase and tumorigenesis in order to provide reference.
