基于UPLC-Triple TOF-MS/MS和多变量统计分析的瓜蒌皮和瓜蒌籽差异化学成分研究

Analysis of Chemical Constituents in Pericarpium Trichosanthis and Semen Trichosanthis Based on UPLC-Triple TOF-MS/MS and Multivariate Statistical Analysis

摘要: 目的:分析瓜蒌皮和瓜蒌籽的差异化学成分。方法: 用超高效液相色谱-串联四级杆飞行时间高分辨质谱(UPLC-Triple TOF-MS/MS)分析技术对同基源瓜蒌皮和瓜蒌籽差异化学成分进行测定。通过二级串联质谱分析,对采集的图谱进行峰匹配、峰对齐、滤噪处理等进行特征峰提取;用主成分分析(PCA)和OPLS-DA分析进行数据处理;根据一级质谱精确质荷比和二级质谱碎片信息,结合软件数据库搜索及相关文献进行成分鉴定。结果:同基源瓜蒌皮和瓜蒌籽化学成分得到有效区别;初步筛选并鉴定出26个差异化学成分,其中20个为共有差异化学成分,并呈现不同的变化规律。结论:从瓜蒌皮和瓜蒌籽化学成分差异的角度,可为揭示二者药性、药效差异的物质基础提供依据,同时也可为瓜蒌皮和瓜蒌籽开发利用提供了科学依据。

Abstract: Objective:To analyze the chemical constituents of pericarpium trichosanthis and semen trichosanthis.Methods:Ultra-performance liquid chromatography-Triple TOF-MS/MS(UPLC-Triple TOF-MS/MS) was used to determine the chemical constituents of pericarpium trichosanthis and semen trichosanthis from the same source. The characteristic peaks were extracted by peak matching, peak alignment and noise filtering through two-stage tandem mass spectrometry analysis. Principal component analysis(PCA) and OPLS-DA analysis were used for data processing. According to the accurate mass charge ratio of the first order mass spectrometry and the fragment information of the second order mass spectrometry, the compositions were identified by software database search and related literature.Results:The chemical constituents of fructus pericarpium trichosanthis and semen trichosanthis were significantly different from those of pericarpium trichosanthis and semen trichosanthis from the same source were effectively distinguished. Twenty-six different chemical components were screened and identified, 20 of which were common and showed different variation patterns. Conclusion:From the perspective of chemical composition difference of pericarpium trichosanthis and semen trichosanthis, it can provide basis for revealing the material basis of drug property and efficacy difference of pericarpium trichosanthis and semen trichosanthis, and also provide scientific basis for the development and utilization of pericarpium trichosanthis and semen trichosanthis.
