HPLC法同时测定“黄芩-黄连”药对中9种成分

Simultaneous Determination of Nine Constituents in Scutellariae Radix and Coptidis Rhizoma by HPLC

摘要:

目的:建立高效液相色谱法同时测定黄芩-黄连中9种成分:黄连碱、表小檗碱、药根碱、小檗碱、巴马汀、黄芩苷、汉黄芩苷、黄芩素和汉黄芩素含量的方法。方法:采用高效液相色谱法,色谱柱为Agilent Zorbax C18色谱柱(4.6 mm×250 mm5 μm),流动相为0.1%甲酸水(B)-甲醇(C),梯度洗脱,流速为1.0 mL·min-1,柱温为30 ℃,检测波长为275 nm,进样量10 μL。结果:黄连碱、表小檗碱、药根碱、小檗碱、巴马汀、黄芩苷、汉黄芩苷、黄芩素和汉黄芩素的线性范围分别为0.73~11.62 μg·mL-1r2=0.999 7)、3.48~55.65 μg·mL-1r2=0.999 7)、2.68~42.88 μg·mL-1r2=0.999 7)、4.23~67.62 μg·mL-1r2=0.999 7)、4.28~68.40 μg·mL-1r2=0.999 7)、15.28~244.40 μg·mL-1r2=0.999 6)、5.74~91.80 μg·mL-1r2=0.999 6)、0.64~10.30 μg·mL-1r2=0.999 6)、0.67~10.64 μg·mL-1r2=0.999 9)。精密度、重复性和稳定性试验RSD均符合要求。黄连碱、表小檗碱、药根碱、小檗碱、巴马汀、黄芩苷、汉黄芩苷、黄芩素和汉黄芩素的加样回收率分别为100.10%100.31%100.02%99.66%100.65%99.83%99.39%100.09%101.31%。结论:本法灵敏简便、准确度高,适用于同时测定黄芩-黄连中9种成分的含量,可用于黄芩-黄连的质量控制。

Abstract:

Objective: A HPLC method was established to determinate Coptisine Epiberberine Jateorhizine Berberine Palmatine Baicalin Wogonoside Baicalein Wogonin in Scutellariae Radix and Coptidis Rhizoma.Methods: Analysis was conducted by HPLC-DAD on Agilent Zorb-ax C18 4.6 mm×250 mm 5 μm column with mobile phase of 0.1% formic acid water B)- methyl alcohol C at the flow rate of 1.0 mL·min-1 by gradient elution. The detection wavelengths were at 275 nm injection volume 10 μL.Results: The linear range of Coptisine Epiberberine Jateorhizine Berberine Palmatine Baicalin Wogonoside Baicalein and Wogonin were 0.73-11.62 μg·mL-1 r2=0.999 7), 3.48-55.65 μg·mL-1 r2=0.999 7), 2.68-42.88 μg·mL-1 r2=0.999 7), 4.23-67.62 μg·mL-1 r2=0.999 7), 4.28-68.40 μg·mL-1 r2=0.999 7), 15.28-244.40 μg·mL-1 r2=0.999 6), 5.74-91.80 μg·mL-1 r2=0.999 6), 0.64-10.30 μg·mL-1 r2=0.999 6), 0.67-10.64 μg·mL-1 r2=0.999 9 respectively. Precision reproducibility and stability RSD all met the requirements. The average recoveries of Coptisine Epiberberine Jateorhizine Berberine Palmatine Baicalin Wogonoside Baicalein and Wogonin 100.10% 100.31% 100.02% 99.66% 100.65% 99.83% 99.39% 100.09% 101.31% respectively.Conclusion: The established method is convenient accurate and suitable for simultaneous determination of 9 ingredients in Scutellariae Radix and Coptidis Rhizoma.
