单细胞测序的技术概述

摘要: 单细胞测序(single cell sequencing, SCS) 技术,是指在单个细胞水平上对其携带的遗传信息进行测序,旨在分子层面获得某种细胞类型的基因序列、转录本、蛋白质及表观遗传学表达谱信息并进行整合分析,目前广泛应用于新物种鉴定、病原筛查、病原进化、发育生物学、神经科学、肿瘤异质性研究以及循环肿瘤细胞等。单细胞测序技术可深层次了解同类细胞不同亚群的分布及状态、作用过程及协作机制等,为探究生命形式和生物行为提供了全新的认识。本文从单细胞测序技术的发展及单细胞分离、多组学的测序等方面作以综述,以期为后续研究提供参考。

Abstract: Single cell sequencing (SCS) technology refers to sequencing the genetic information at a single cell level, which aims to obtain and analyze the gene sequences, transcripts, proteins and epigenetic expression profile information of a specific cell type. It is widely used in new species identification, pathogen screening and evolution, developmental biology, neuroscience, tumor heterogeneity research and circulating tumor cells at present. SCS technology can make us gain a deeper understanding of different subgroups of the same type of cells, and provide a new understanding of exploring life forms and biological behaviors of organisms.Here, we conducted a brief overview of the development of SCS, single-cell isolation and multi-omics, and summarized their progression, to provide a reference for the further researches.
