地黄配方颗粒指纹图谱研究

摘要: 目的:建立地黄配方颗粒的高效液相指纹图谱,为其质量研究评价提供实验依据。 方法:地黄配方颗粒样品经水超声溶解后,通过D101型大孔吸附树脂柱洗脱,洗脱液蒸干溶解于乙腈-0.1%磷酸,采用Welch Ultimate LP -C18(4.6 mm×250 mm,5 μm)色谱柱,以乙腈(A)-0.1%磷酸(B)为流动相,梯度洗脱,检测波长330 nm,体积流量1.0 mL·min-1,柱温30 ℃,进样量20 μL,记录时间90 min。结果:地黄配方颗粒精密度、稳定性和重复性试验的RSD分别为0.03%、0.06%、0.21%,均不大于2.0%(n=6)。采用梯度洗脱得到的色谱图各色谱峰分离效果好,达到指纹图谱的要求。与地黄药材、饮片及汤剂的指纹图谱进行系统比较,结果发现各批次样品的相似度较高,均一性较好,均呈现出8个共有峰,各共有峰相对保留时间稳定,具有专属性。结论:该方法准确可靠,重复性好,可为地黄配方颗粒质量评价提供参考。

Abstract: Objecetive: To establish an HPLC fingerprint of Rehmannia glutinosa Libosch. dispensing granules and provide experimental evidence for its quality criteria. Methods: Take water extract of Rehmannia glutinosa Libosch. dispensing granules by ultrasonic processing and then eluted by D101 type macroporous adsorption resin column. The evaporation of eluent was dissolved by acetonitrile-0.1% phosphoric acid. HPLC analysis was carried out on Welch Ultimate LP -C18(4.6 mm×250 mm, 5 μm)chromatographic column with mobile phase of acetonitrile(A)-0.1% phosphoric acid(B), gradient elution with a flow at 1.0 mL·min-1, an ultraviolet detection wavelength at 330 nm. The column temperature was 30 ℃, the injection volume was 20 μL and the record time was 90 min. Results: The RSD of the precision, stability and repeatability test of Rehmannia glutinosa Libosch. dispensing granules 0.03%, 0.06%, and 0.21% respectively, all of which were not more than 2.0% (n=6). The separation effect of each chromatographic peak of the chromatogram obtained by gradient elution was good, which meets the requirements of fingerprint. A systematic comparison with the fingerprints of Rehmannia glutinosa medicinal materials, decoction pieces and decoctions showed that the similarity of each batch of samples was high, and the uniformity was good. They all showed 8 common peaks. The relative retention time of each common peak was stable and exclusive. Conclution: The method is accurate, reliable, and reproducible. It can provide a reference for the quality evaluation of Rehmannia glutinosa Libosch. dispensing granules.
