柴术理胃饮对肝郁脾虚型功能性消化不良大鼠胃肠运动及血清SP、VIP含量的影响

The Effect of Chaizhu Liwei Yin on Gastrointestinal Motility and Serum SP and VIP Contents in Functional Dyspepsia Rats with Stagnation of Liver Qi and Spleen Deficiency

摘要: 目的:研究柴术理胃饮对造模后功能性消化不良(functional dyspepsia,FD)大鼠的胃肠运动、胃组织病理学表现和血清P物质(substance P,SP)、血管活性肠肽(vasoactive intestinal peptide,VIP)含量的影响。方法:SPF级健康雄性SD大鼠采用复合因素刺激法(慢性疲劳+悬尾+饮食失节)制作FD模型,随机分为模型组、柴术理胃饮低、中、高剂量组和多潘立酮组,各组分别设置10只,另设10只未造模大鼠为空白组。各组给予相应的药物后观察大鼠的饮水量、进食量、体重等一般状态,根据公式计算胃排空率和小肠推进率,大鼠血清SP和VIP的含量选取ELISA法检测,胃组织病理学的变化选取HE染色法观察。结果:治疗后,柴术理胃饮各剂量组大鼠胃排空率与模型组相比上升,小肠推进率与模型组相比提高(P<0.05);模型组大鼠血清中SP含量与空白组相比降低(P<0.01),VIP含量与空白组相比升高(P<0.01);柴术理胃饮各剂量组大鼠血清中SP含量与模型组相比均升高,VIP含量与模型组相比均降低(P<0.05)。结论:柴术理胃饮可以改善肝郁脾虚型FD大鼠的胃排空功能和小肠推进功能,具有提高血清SP含量和降低血清VIP含量的作用,这或许是此方治疗FD的机理之一。

Abstract: Objective: To study the effects of chaizhu liwei yin on gastrointestinal motility, gastric histopathology, serum level of substance P (SP) and vasoactive intestinal peptide (VIP) of rats with functional dyspepsia (FD). Methods: SPF healthy male SD rats were stimulated by compound factors (chronic fatigue + tail suspension + diet defect) to make FD model. They were randomly divided into the model group, chaizhu liwei yin low dose group, middle dose group, high dose group and the domperidone group, with 10 rats in each group, and another 10 rats without making FD model were set as the blank group. After the corresponding drugs were given to each group, the general situation of the rats, such as water intake, food intake and weight were observed. The gastric emptying rate and small intestinal propulsion rate were calculated according to the formula. The contents of SP and VIP in serum were detected by ELISA, and the changes of gastric histopathology were observed by HE staining. Results: After treatment, compared with the model group, the gastric emptying rate and small intestinal propulsion rate of rats in chaizhu liwei yin each dose group were higher(P<0.05). The serum SP content of rats in model group was lower than that of the blank group and the serum VIP content was higher than that of the blank group(P<0.01). Compared with the model group, the serum SP content of rats in chaizhu liwei yin each dose group increased, and serum VIP content of rats in chaizhu liwei yin each dose group decreased (P<0.05). Conclusion: Chaizhu liwei yin can improve the gastric emptying function and small intestine propulsion function of FD rats with stagnation of liver qi and spleen deficiency, and can increase serum content of SP and reduce serum content of VIP, which may be one of the mechanisms of chaizhu liwei yin treating FD.
