响应面法优化复方柏连灌肠剂的提取工艺研究

Study on Optimization of Extraction Process of Compound Bailian Enema by Response Surface Method

摘要: 目的:优化复方柏连灌肠剂的提取工艺,为申报医疗机构制剂提供参考依据。方法:建立复方柏连灌肠剂中没食子酸的高效液相色谱(HPLC)测定方法,以没食子酸转移率为评价指标,在单因素试验基础上,应用 Box-Behnken响应面中心组合法进行因素水平设计,考察料液比、提取时间和提取次数对处方中没食子酸提取效果的影响。结果:本研究建立的高效液相色谱分析方法的专属性、精密度、重复性、稳定性均符合《中华人民共和国药典》要求,没食子酸在24~144 μg·mL-1范围内进样浓度与色谱峰面积呈现良好的线性关系,回归方程为:y=27.794x+44.413,r2 =0.999 9,平均加样回收率为99.6%(RSD=2.7%)。复方柏连灌肠剂最佳提取工艺条件为提取3次,每次加10倍量水,每次1 h。实测与响应面法回归模型高度拟合。结论: 本研究通过响应面法优选的复方柏连灌肠剂提取工艺稳定可靠,质量可控。

Abstract: Objective:To optimize the extraction process of compound bailian enema and to provide a reference for the application of preparations for medical institutions. Methods: Establish a HPLC method for the determination of gallic acid in compound bailian enema. The transfer rate of gallic acid was used as an evaluation indicator.Based on single factor experiments,the Box-Behnken response surface center combination method was used to design the factor level and investigate the effect of solid-liquid ratio, extraction time and extraction times on the extraction effect of gallic acid in the prescription.Results:The specificity, precision, repeatability, and stability of the established high performance liquid chromatography method met the requirements of the pharmacopoeia. The injection concentration of gallic acid in the range of 24-144 μg·mL-1 showed a good linear relationship with the chromatographic peak area. The regression equation is: y=27.794x+44.413, r2=0.999 9, and the average sample recovery rate is 99.6% (RSD=2.7%). The optimal extraction conditions for compound bailian enema are as follows extrant 3 times. 10 times the amount of water each time,extract for 1 h each time. The actual measurement and the response surface method regression model are highly fitted. Conclusion: The extraction process of compound bailian enema established in this study, which was optimized by response surface method is stable and reliable, and the quality is controllable.
