黄连降低耐甲氧西林金黄色葡萄球菌耐药性的作用机理研究

摘要: 目的:探讨黄连原液降低耐甲氧西林金黄色葡萄球菌(MRSA)耐药性的作用机理。方法:将10株耐甲氧西林金黄色葡萄球菌USA 300根据黄连原液浓度不同,分为黄连原液50.00 μg·mL-1组、25.00 μg·mL-1组、12.25 μg·mL-1组、6.25 μg·mL-1组、3.12 μg·mL-1组、1.56 μg·mL-1组、0.78 μg·mL-1组、0.39 μg·mL-1组,同时设阴性对照(只含营养肉汤管)。使用结晶紫法、Western blot、CCK-8法和RT-qPCR法检测黄连原液通过清除、抑制USA 300生物膜形成所发挥的抑菌作用。结果:黄连原液抑制USA 300的最小抑菌浓度(MIC)为6.25 μg·mL-1;随着黄连原液浓度的增加,黄连原液对USA 300的抑制率、清除率、抑菌圈、逆转率均升高,相关基因和蛋白表达均降低 。结论:黄连原液通过抑制USA 300生物膜形成、抑制生物膜icaA、atlE、aap、agrA和stk蛋白及mRNA的表达,加大对USA 300生物膜的清除率和抑制USA 300的活性,降低USA 300的耐药性。

Abstract: Objective: To investigate the mechanism of Coptis Chinensis solution to reduce drug resistance of methicillin resistant staphylococcus aureus (MRSA).Methods:10 strains of MRSA USA 300 were tested according to the concentration of Coptis Chinensis solution,divided into the 50.00 μg·mL-1group, 25.00 μg·mL-1 group, 12.25 μg·mL-1 group, 6.25 μg·mL-1 group, 3.12 μg·mL-1 group, 1.56 μg·mL-1 group, 0.78 μg·mL-1 group, 0.39 μg·mL-1 group, the negative control (containing only nutrient broth tube) was set at the same time. Crystal violet method, Western blot, CCK-8 method and RT-qPCR method were used to detect the bacteriostasis of Coptis chinensis solution by removing and inhibiting USA 300 forming biofilm.Results:The MIC of Coptis Chinensis solution inhibition of MRSA USA 300 was 6.25 μg·mL-1. With the increase of Coptis Chinensis solution concentration, the USA 300 inhibition rate, clearance rate, inhibition circle, reversal rate were all increased, and the expression of related genes and proteins were all decreased. Conclusion: By inhibiting USA 300 forming biofilm, inhibit the expression of icaA, atlE, aap, agrA and stk proteins and mRNA in biofilm. Coptis Chinensis solution can increase clearance of USA 300 biofilms and inhibite USA 300 activity,reduce the drug resistance of USA 300.
