基于网络药理学的中医药治疗不明原因复发性流产的作用机制分析

摘要: 目的:探究中医药治疗不明原因复发性流产(unexplained recurrent spontaneous abortion,URSA)的用药规律,预测核心药物的潜在作用机制,以期为中医药治疗URSA提供理论依据和参考。方法:检索中英文数据库中关于中医药治疗URSA的相关文献,构建URSA处方数据库,运用Excel 2019软件对方剂中的中药进行用药频次统计分析,运用SPSS Modeler 18.0软件进行复杂网络、关联规则分析,运用SPSS Statistics 27.0软件进行聚类分析,运用Cytoscape 3.9.1软件进行网络可视化分析;借助TCMSP数据库获取核心药物组合的活性成分和靶点,通过GeneCards等数据库预测URSA的疾病靶点,利用STRING 11.5数据库和Cytoscape 3.9.1构建PPI网络图,并筛选关键靶点,借助Metascape数据库进行GO功能和KEGG通路富集分析。结果:数据库共筛选出30首中药方剂,涉及63味中药,核心药物组合为菟丝子-续断。核心药物与疾病的交集靶点80个,关键靶点21个,包括VEGFA、TP53、AKT1等。GO功能和KEGG通路富集分析发现,共同靶点主要涉及流体剪切力与血管粥样硬化、癌症通路、糖尿病并发症中的AGE-RAGE等多种信号通路。结论:中医治疗URSA以补肾健脾,调补冲任为主,其核心药物组合通过多成分、多靶点、多通路治疗URSA,为中医药治疗URSA提供了理论依据。

Abstract: Objective: To explore the medication rules of traditional Chinese medicine (TCM) in the treatment of unexplained recurrent spontaneous abortion (URSA) and predict the potential action mechanism of core drugs, in order to provide theoretical basis and reference for the treatment of URSA with TCM. Methods: The literatures on TCM treatment of URSA were searched in Chinese and English databases to construct URSA prescription database. The Excel 2019 software was used to make statistical analysis of the use frequency of Chinese herbs in the prescriptions.SPSS Modeler 18.0 software was used to analyze complex network and association rules. SPSS Statistics 27.0 software was used for cluster analysis, and Cytoscape 3.9.1 software was used for network visualization analysis. The active ingredients and targets of the core drug groups were accessed from the TCMSP database, and URSA disease targets were predicted by GeneCards and other databases. STRING 11.5 database and Cytoscape 3.9.1 were used to construct the PPI network map, and screen the key targets. The Metascape database was used for GO function and KEGG pathway enrichment analysis. Results: A total of 30 TCM prescriptions were screened out, involving 63 Chinese herbs, and the core drug group was dodder-dipsacus. There were 80 intersection targets between core drugs and diseases, and 21 key targets, including VEGFA, TP53, AKT1, etc. GO function and KEGG pathway enrichment analysis showed that the common targets involved a variety of signaling pathways such as fluid shear stress and vascular atherosclerosis, cancer pathway, and AGE-RAGE in diabetic complications. Conclusion: TCM treatment of URSA mainly focuses on tonifing the kidney and spleen, regulating and tonifying Chong-Ren, and TCM core drug group treats URSA through multi-component, multi-target and multi-pathway, which provide a theoretical basis for the TCM treatment of URSA.
